That's awesome thanks guys. Sheer amount of LC was likely reason for contam as all previous transfers have been fine in same conditions (SAB, cleaned room, cleaned body, bleached workspace/tools, face mask, hair tied back, sealed windows/doors). Is there a way to actually check that the LC is viable and contaminant free past simply looking at it?

Like play it "castles in the sky" and if it doesn't do a jig then it's obviously rotten.

Wow!  Thanks y'all for the excellent discussion about condensation in plates. It seems there really ARE 1000 ways to skin a cat. So may good ideas. One thing is clear...mitigating strategies are less risky in front of a hood...FB's wipe-down, for example. I'm still working in a SAB for now, and Iove pouring my own plates - it's like meditation for me - and I'm thinking that simply replacing the wet lid with a sterile dry one could work with hood or box. And I will absolutely start to store vertical, because it's fucking cool. I'm a ClingWrap guy so that should work.  See next post for another question that has come out of this....

@Arky, that grow room is gorgeous. I've seen a few pix of rooms like that and I always drool.

I have to ask...what's with the tinfoil wall coverings?  How is this accomplished and is there evidence that it cunts down on contam?  I assume it greatly improves temp control...perhaps there is a room Tek or diy build log I have discovered yet?

Thank you. It's a 5x9 Gorilla grow tent that I’ve set up inside a finished room in my basement. While my initial intention was to minimize contamination—similar to a laboratory environment using a fan-filter unit (FFU)—I’ve found out it functions more like a large Martha tent, helping me better control temperature and humidity. Instead of reducing contamination, it essentially was a still-air box (SAB) with a fan that would give me fits with contamination while working right in front of the fan. I had to remove the FFU and table to getter better results with my FFU (thanks to sage advise I received from fellow members here on the Shroomery!).

This setup is quite effective for maintaining a stable environment for my tubs, jars, and cultures, keeping the temperature around 75°F. I achieve this with a radiator heater, while a homemade 45-liter humidifier helps maintain humidity levels between 40% and 60% relative humidity (Rh). Additionally, I have a HEPA-filtered intake fan that brings in fresh air, creating positive pressure that pushes the air out through the back of the tent.

Charlie Brown germ plate looking good

This is a Green Cap Ochra germ plate. I have another ochra culture going, but I'm not sure if it's a clone or from a print. Not sure if black cap or blue cap either lol. I have another germ plate in case this one is off, but the large piece looks like mycelium to my eye, maybe not so much in the photo.

Made some agar recently without realizing I didn't have enough cups left. I used a few that had been out of the sleeve or touched the bottom of the SAB, normally those are left to the side for non agar purposes. Since I was really low, I decided to use them. It's been over a week and no sign of contamination, so I will use them.

And this isn't quite agar, but rather , a scoby like mass that grew on top of a LC that I never tried using. First pic is from 2 months ago

Gotta say, I feel the urge to test it out even though it's somewhat sketchy. Yeti x Husky hybrid

Awesome, Arky. Thanks for the break down. So where is your FFU located now - where do you do your sterile at agar work etc?

Oh since we're doing a lab share and I happen to have plates in front of my ffu.

Got everything setup and reorganized while on break, feels a lot less stifling.

I would love to get more art in here though. Maybe make a collage out of cards and stickers folks share with me

Okay that's it! I'm not fucking around anymore. I've got an old wine closet which I don't need anymore because I've lost all desire to drink alcohol. This is my official notice that I am converting that room to the sickest lab imaginable. It'll probably take me 6 months of negotiating with the wife but it's going to happen.

That is all

Framed spore prints always feel like they go hard hanging in a lab space…. If you’re thinking of art.

I can't tell if the one on the left has contamination but I think so. It looks like mycelium from a chestnut mushroom but I think its some kinda mold. No way it had that much growth starting from a spot that I can't visibly see spores. I think the one on the right is looking ok.

They South African Transki that's what this is supposed to be its a strange looking Transki I can't wait to see what it looks like

Oh also that's the backside of the plate to much condensation to take a good picture of the other side. It definitely looks like a different type of growth than the plate on right.

Ок i start to strain my no pour brf agar and finally start to noticed my problems. So i can see clearly now and noticed small bacterial spots here and there in some plates. I think biggest part of the problem i cant clean my cultures is water in no pour agar. Bacteria spread very fast from water and if i have contaminated culture from start is very hard to clean because of exess of water. Even when i try to select good transfer if there is bacterial spot far away im sure with that condensation bacteria will jump on my new plate. I start to dump exess water in Sab. And maybe will need to made a lot of agar plates and waiting weeks for all condensation in plates to evapore or be absorbet on agar. Any other advice what else i can do.

Some good looking to me clean agar

And condensation on lid.

Here i try to cath bacterial satelite some mycelium that dont look very good and water all around my plate

First plate looks good! If your plates are clean, condensation isn't a problem anyway.  But it makes sense to do what you say: make a lot more plates and wait before using. You can leave them unwrapped a while, you can store them upside down, you can put a plate of warm water on top of a stack to help with condensation. Get it!
