Wouldn't sterilization matter?

If not I am happy to forget it.

If you were to colonize it sealed like in a spawn bag or a jar. And inoculate it in a sab or flowhood, then it could work.

If you are sterilizing it, then dumping it on coir open air it will mold before it gets colonized

Does this transfer look isolated? This is on lme regular recipe agar. Next round of agar I'll go half lme to try to get heartier growth.

It is definitely isolated from the edges of that plate.

Maybe monoculture would be a better term. I'm just going to clone something from the next good pinset. I was trying to get fast rhizomorphic growth and that transfer is lacking that.

dont worry about that.  just grab a good clone and run it.  eyes on the prize.

Not a better term.

I think of a monoculture in this instance as being a monokaryon versus a dikaryon.

And if you don't know what those are... a quick google search can tell you everything you didn't know you wanted to know about it :super:

I would say that it appears you've isolated the mycelium you wanted away from contamination.

And remember, rhizo growth isn't the be all and end all of culture work. People romanticize it too much

Eyes on the prize, and your hand on your balls. Pick one,the hand doesn't matter.

Both isolates and monocultures arent very useful for most of us

Rustywhyte is another culture that is usually tommy but I've had rhizo cultures too

One of the best cultures I had looked like mold wasn't even tomentose really. Can't remember if it was a rustyhwyte clone or something else

it really makes no difference.  rhizo cultures are just more photogenic.

Roger that, rhizo growth was all the rage a decade ago. I'm just returning after a five year hiatus, but both of my comeback boxes that recently fruited were tomentose.

Ive gotten a mold that looked close to tomentose growth until it aged and turned more gray in color. Saving grace was it popped up on the edge of the plate, so was immediately suspicious. It still makes me nervous that someday that could be enmeshed with mycelium and Id struggle to spot it. Ropes give me more confidence for now.

Gandalf germ plate from spore swab inoc on 8/8/25

x7x+ T1 plate 8/12/25

Is this thickness normal?

How much DME(dry malt extract)?

Oof, wish I knew.  I'm using the premade LME from Rak Media.  The sediment in the gandalf plate is due to me not mixing the it good enough before sterilization.  The second time I made plates, I was much more thorough during mixing and had zero sediment.

Yeah.

Wanna save some money on pre mixed media?

Grab some dry malt extract and some agar powder... mix the two at 1:1 ratio. Now just use 20g of powder per 500ml of water.
