Got it. Thanks a lot for sharing your knowledge. I do suck out as much air as possible but sometimes since I use double ziplocks for safety so if a bag breaks it doesn't mess with the moisture content. I have seen some small air pockets.

I did another pasteurization run last night and 3 times at different times I took one bag out to measure the temp inside. I use pretty small bags with 3 quart of substrate inside and the core did get up to correct temp:shrug:

I will start to use my vacuum sealer and stop using ziplocks and double bags though.

Again thanks! I will come back in a week with a report because I did spawn 3 jars to bags that was pasteurized exactly the same way as the bags I had the contams in. So we will find out if it was GE or improper pasteurization that was at fault.

Gonna be more careful and use substrates ASAP in the future either way.

That being said I have my last 2 jars to spawn right now:leocheers:

Normally I use foil on top and poke a few small holes. But wanted to cut down on the amount of foil I use, so one of the trays I will put the lid on but without clamping it down obviously as a test:takingnotes:

Obviously ppl have been pasteurizing without vac sealing with success for a very long time so it’s not an all or nothing sort of deal but it’s one less thing you need to worry about.. I have zero doubt I’m reaching temp with the VS.  it also helps equalize the water distribution in you substrate

First of an update on my post about few days ago. It's been awhile and cased all of the trays 3 days back. All looking good so far.

So I'm pretty certain the problem with my substrate getting the white contams was the fact that I left them in a closed bag and didn't give them any air.

Got another question now though:shrug:

Posted in the agar thread but didn't get anyone answering so thought I try here.

I do like the way the Rancho Agosto looks a lot more than the Tampa.
Is it normal for APE to be this thick? It grows very fast so I'm happy with that. But haven't had such thick growth in a long time. Normal or is there a reason for it?

Has anyone ever tried to run a plant mister off of an aquarium pump? I'm trying to figure out a way to automate my misting for my pans and semps

1.5% LME and 2% agar so very normal I would say:shrug:
Well I will see what it looks like once I transfer to agar tomorrow.

Good to know it's nothing to worry about. Thought maybe APE looked a bit different.

Someone please help me. This been driving me insane.

I had strange looking growth so didn't transfers to BRF pucks. Growth looked much better, at least to me. Now I did transfers to new plates 7g mea 10g agar per 500 ml.

Same looking growth again. I did open a plate and the agar felt very hard. Only thing I could think would be a got new agar powder. Could it be that my new agar powder is making my plates to hard and the mycelium is having a hard time colonizing the plates?

No specifics, sometimes cultures have stuff riding along that normal transfers cant take get rid of. Whats the recipe of that agar?
I would try cabin sequestering or a hot pour and start more spores in the meantime

Yeah I hear you. Those plates are transfered from BRF pucks though that I thought would clean them up. Guess I will give water agar a try and trenching as well to see if that helps:shrug:

Just strange that all the cultures I got going right now has the same problem..

Recipe is  7g mea 10g agar per 500 ml

Yeah that definitely looks like ridealong bacteria interacting with the myc to me in top 2 rows.  The pucks just look like typical puck myc from uneven distribution of nutrition inherent to pucks. 
A single bad session can distribute a contaminate across several plates.  I hate it when that happens.

How do you guys prepare slides for observing mycelium?

I want to look at some dikaryons to practice finding clamps because my goals to source monos from what I got going on currently.

Do I put a slide cover over the mycelium and squish it?
I’m not sure if it’s gonna obstruct my view and I have limited materials currently.

Is there specific ways to cut to not mangle up the mycelium?

Is a smaller piece better? Or should I use a bigger piece? I was thinking grain of rice, but I’m not sure if that area would be sufficient to confirm a mono?

People have different opinions on this.  Some say use a piece of transparent tape to touch the mycelium and that will take a suitably thin layer to apply to a slide.  I've seen people say "use an inoculation loop to grab a sample."  Some people just grab a 1-2mm piece from a dish.  It's finicky.  The tape thing works good though.  Old school.  That said, I'm essentially a useful idiot.  Hopefully Supa or Nichrome or somebody else with the appropriate experience will chime in.

Yeah I tried cutting and squishing and it was really hard to see wtf was going on too much agar I think.

Does manure based substrate that's been opened and sat for 2-3 weeks need to be re-pasturized? Sub mix is grass, manure. and CVG.

This tub of purple mystics (from sporeworks LC) seemed healthy all the way through, but some condensation pooled in the middle of the cake that I didn't catch quickly enough so the myc matted a bit in the middle before I got rid of it. It flushed a bit but not great, I got a couple dried ozs of smaller fruit from it. Had a lot of fuzzy feet and some condensation had pooled in the bottom of the tub, so I tried to up the airflow a bit in the monotub and also dumped out all the collected condensate. But I had to leave town for a few days, and came back to this sad-seeming sight. This is only my second tub, first one I knew the spawn was bacterial but sent anyhow, so I don't have a lot of experience recognizing what's wrong and whether it's fixable. The spots without myc are just where I harvested some of the healthier-looking fruits with torn veils. Any hope to get more out of this?

Whats the sad sight? Aborts? Eat em and rehydrate.

How does it smell? Theoretically if it was allowed to breathe it should be fine ifnit was properly pasteurized

Are they aborts? It's clearly not from lack of moisture... Is the white fuzz growing over the whole fruits a mold? Is there anything I should do differently based on the conditions you see in the picture? I was worried that attempting to rehydrate and go for flush #2 would let whatever might be questionable in that tub to spiral out of control.

I'm just trying to figure out how to get those healthy-looking canopies of monster fruits I see all over the forums :smile:
