I only run LC, and I mist it!

But I have been doing some quick n dirty in the plate LIs, works really well. I usually score up the surface with a scalpel before adding the water and shaking. A couple of the plates have actually pinned AFTER being made into LI!

In fact, that might be where the gold spore PE came from.....

Also Piggy I suppose I do largely agree. I think the difference in my approach vs convention is I’d put it earlier in the workflow as an option, and I think it has a little more utility to the hobbyist than it’s given credit.

Otherwise I guess my take wasn’t as scorching hot as I thought lol

What do you mean by earlier in the workflow?  Once someone is competent working with agar they are ready seems pretty asap to me

As for usefulness, yes it can absolutely have uses in home cult.  Again I'm just usually trying to get people to crawl before they run.

I’m more of a momma bird pushing babies out of the nest and scooping them up if they can’t fly kinda guy.

I think, as long as you’re not using overtly bad inoculant like spores, it’s ok to learn agar and LC together. Yeah you need clean agar to make LC, but sometimes the LC can even tell you when your agar wasn’t as clean as you thought. I guess I don’t separate the two. If I were teaching a class, agar would be a lesson before LC but they’d be on the same test.

Wait, you test your liquid cultures?

In all seriousness, if everyone tested LC before using it, the risks for new growers would be a lot lower. But if you don't know how to use agar in the first place, you're sure not going to be able to test your LC (you said it better than me) - and if you're doing it because you want to shave off some time, you're probably not going to bother testing in any case...

I fully admit to almost never testing my own LC before using it. But I also am pretty confident the tiny piece I used to inoculate it was axenic, and failing that, I know what to look for in liquid. I don't think that's as straightforward as recognizing contamination on a plate, though, especially if you don't have enough experience to know what good growth looks like.

We test LC like we test code - in production.

Plin  clone. Does this look ok to you guys? There is a slight translucent band just outside the mycelium. Am I just tripping out?

I tested my first LC on agar and then tests were performed on a grain jar after that.

Im from around the way - I'm leavin here with somethin.

People often say "but it tested clean", only for grain to contaminate. I wonder what percentage of that is contamination from handling while testing.

I considered something along those lines when my last LC "test jar" shit the bed. It's hard for me to believe I screwed up a tiny wedge to a bottle so I was left with wondering if I introduced the bacteria when I inoculated the jar. I suppose the test can prove it's clean but can't prove that it's unclean...whatdoyado...other than run like 3 test jars, which I'm not gonna do. Ill just make new LC.

Speaking of making new LC - I wanna toss sus LC back in the PC as some peak laziness shortcut. What are your thoughts on that stipe? Everything gonna die for sure, but would you still respect me in the morning?

If you are doing an agar to grain transfer I would recommend letting the entire plate fully colonize if possible.  Did the plate just go from consistent growth and just slowed down out of no where?  Clearly your plate is an agar to agar transfer so I would have to assume this isn't your first transfer go around with this strain.  Is this the only recipient plate you inoculated from the source/donor plate?  Do you have any other donor plates?  Give me some more details and I'm sure i can be of some help.

Whatever you are doing keep it up!  The purity of the rhizomorphic growth you're getting from a grain to agar transfer really shows the strength of the genetics you're working with.  Absolutely beautiful!!

I'm not very familiar with the HBP strain but just wondering what the inoculation source was?  I would have to imagine LC inoculation?

Please elaborate with exactly you have going on here.  Is that growing on an agar plate containing only water and agar agar???  Was the transfer source colonized grain?

Point taken, and I've edited "commercial grows" out of my
"FROM SPORES TO TRIPS" post, now says

--Liquid Culture (LC). Don't do it until you master clean agar plates.

LC is unneeded unless you're need to grow a lot (or just want to do try something) If not, why not just toss agar slices into grain jars, and shake at 30 percent colonization? You're much more likely early on to get clean grows.....

But I am still going to recommend new growers not do LC until they've mastered clean plates, spawn, transfers, recognizing contam, etc.

Thanks

Welcome to the Shroomery.

No idea who this response was meant for, but I assume not mushboy. Curious though why you think it's important let a plate fully grow out before sending to grain.

By the way, fewer replies and more quoting makes for easier to follow threads (see the "quote" link next to the reply button).
