I just looked at cryovials. The 5ml. Not sure how much they hold, but it has to be better storing agar a samples from clean plates in those in the refrigerator than a bunch of plates I would assume? I am about runneth over with plates ATM and hate to toss some of these as they are really pretty and clean.

For the same reasons I'd suggest a new grower not try to "experiment" until they've grown with proven teks and got them down. Cart before horse. 

People will do what they're going to do, and LC seems fun, but it seems to result in contam with new growers, or new growers not sure if they have contam, or just tossing spores into broth.

I think you and I have had this discussion before. It not, it was something similar.

I'll keep suggesting people don't do it at first and you keep questioning it.

You don't even need to store them in the fridge. Just put distilled water in them, sterilize, and add a few pieces of colonized agar. You can keep them at room temperature.

I use the 2ml, for which you can also find convenient 100-spot storage boxes on Amazon. The 5ml ones will work just fine too.

They're discussing the pitfalls/necessity of LC.

Storing cultures in distilled water is groovy af all the time.

I'm with you on discouraging new growers from trying to use LC as a shortcut - notably syringe to broth is asking for trouble, even with LC, and unfortunately there are a number of vendors advertising exactly that: buy this LC syringe and expand it straight into to a much larger amount of LC.

I think it's a little over-simplistic to say it's only useful for commercial growing. It's certainly a tool that allows inoculating a lot more spawn with less work compared to growing out plates. But it can also be used for one spawn jar at a time. One of the things I use it for is to have bottles ready of cultures I like to grow frequently, and it avoids some just-in-time agar work - I can go from the idea to grow something to having an inoculated grain jar in one day (or 5 minutes if I already have sterilized grain). It stores really well, especially if the nutrient level is low enough that it doesn't try to make a mycelial mat on the surface.

I'm experimenting with ReverendMyc's stored LI for the same purpose - replacing the LC colonization step with agar plate colonization is appealing.

Anyway, I think a better principle might be something like "Don't try making LC before you can consistently get (and recognize!) clean culture on agar", and never as a shortcut on an early grow.

Go to that link and you get "Liquid Culture (LC). Don't do it until you master clean agar plates....why not just toss agar slices into grain jars, and shake at 30 percent? You're much more likely early on to get clean grows.

If you must do LC, Never add spores to LC broth. Always grow spores or clone first on agar, make sure it's clean, then send a TINY piece to the LC.

LC can be a great way to grow a lot of shrooms fast, but it's also a great way to grow a lot of mold and bacteria fast."

That's my take on LC as well.  A more abstract way to think of it is master 2D before you go 3D.

Speaking of pimple popping tools, I just wanted to share a bit about my current GT project.

Golden Teacher was my very first culture years ago, and it still has a nostalgic place for me, learned PF Tek with it and was immediately hooked. Unfortunately, I contaminated my original spore print at some point, so I recently ordered a fresh GT syringe to start over clean.

This time around, I’m working with BRF pucks and have been experimenting with different tools for transfers. For the first and second plate i used tweezers, and for the third that dental pick-style tool that comes in a pimple popping kit. Honestly, I really like it, it's super precise and great for taking ultra small samples.

Being able to store for a really long time at room temp is a pretty big plus.

I fully understand that I am in an extreme minority here, but I just want to say LC and SSLC are identically easy to inoculating grain. If you can put a wedge of agar to grain, you can put a wedge of agar to a bottle of juice. In fact, it’s much much easier to make the broth for SSLC than it is to make good grains.

Insofar as using it, it’s even easier than agar to grain. Just pour a little sauce in your nugget box and you are rocking and rolling.

SSLC isn’t just a tool for bulk growing, but a great way to explore the phenos hidden inside a culture. As Piggy said, it’s 3D agar, essentially. So where a plate sectors in 2 dimensions, SSLC (or just regular LC) expands in 3. Globes instead of circles.

I understand to a degree why we steer people away from it, but really if you can do a clean agar transfer to a new plate you’re already doing more complex movements than LC. I think liquid culture and agar can honestly be learned in tandem. I know that’s what I did, and I’m not special or more adept than any other new grower here.

It’s just a bottle of broth. Genies don’t come out when you open them or anything. A2G is the same thing as A2broth, but the broth is easier to prep.

Ok. Eviscerate me now. I am ready.

I mean I don't see you disagreeing with what most experienced people are saying about LC, SSLC, ECT.  In the scenario you have presented the cultivator can already develop and identify a healthy clean culture on agar to transfer into LC.  By that point yes, they are ready for LC.  So, in essence, you have agreed with what most people are already saying and are not an outlier on the issue.

I think what most people are saying is there's this new craze about LC and newbies are trying to jump right into it without any experience or understanding what they are doing, often with skipping agar altogether.

For just a second I had an issue with the more phenos in 3d than 2d as it's the exact same thing just flat or globular, but then my brain did the rest of the math and that globe broken up into an LC is going to have more phenos than a tiny agar wedge taken from a plate.

TLDR: I think you're mostly agreeing with everyone :shrug:

If you're gonna get jumped, I can't just stand by and watch like some fuggin slouch!

I 100% agree and basically said the same thing to someone a while back. This isn't objective fact, but it works for me and the hiccups I'm prone to. Slicing a plate into quads and transferring to a jar sketches me out way more than a 2 second pour from a bottle.

Edit: As stated, being able to identify a clean plate is 100% necessary. Which is pretty well true for making clean spawn anyway, but if you can't recognize a clean plate, you can't reliably test the LC, which then means you're going to grenade more spawn than if doing a2g with 1 or 2 jars.

Also Piggy I suppose I do largely agree. I think the difference in my approach vs convention is I’d put it earlier in the workflow as an option, and I think it has a little more utility to the hobbyist than it’s given credit.

Otherwise I guess my take wasn’t as scorching hot as I thought lol
