Does anyone else's transfers go like this:

Step 1. Look at all your plates and select the one with the best growth
Step 2. Select the sector with the most beautiful rhizomorphic growth
Step 3. Carefully cut out a tiny piece to transfer
Step 4. Accidentally drop that piece onto the floor of your SAB
Step 5. Convince yourself that the piece you selected probably had shitty genetics and your new piece will be amazing

Or is it just me?

There seems to always be a part of the process where a nice transfer slides off the blade in any project I do

I know there’s some issues with the plate, but beyond that - this is T2 of TTBVI from a spore print.

I have no experience with pans really. I read it’s fast and wispy, but this looks like mold to me.  Thanks for any opinions.

I'm attempting to make up my first agar plates. Attempt #1 was a fail. Attempt #2, last night I mixed up 400ml of agar using 8g of LME and 8g of agar. It's mixed well and I put in the refrigerator. It's nice and clear but I still need to sterilize it. I should first remelt it, correct? Then into PC at 15 PSI for 30 minutes?

Thin, slow, and erratic growth. Some cultures won’t grow at all, they just sit on the plate and do nothing.



I just experienced something similar. At first I thought it was the blue food colouring, but I’m not sure anymore. It's better on the plates without the colouring, but still bad.




Got them over on BRF(1,5%)/ELME(0,5%) plates, with the following results thus far:




My theory is that either higher temps have influenced the way they grow on the ELME or it's gone bad.

Unfortunately I don't possess constant and wide enough data for any comparison, so it's nothing but speculation from my side. Higher nute content, different agar stiffness. The only thing the bad ones have in common is 1-1,6% ELME.

The only thing I know to be true, is that all my cultures do better on BRF/ELME instead of just ELME.

I have the same water source, using the same filter all across the board.
The water is pretty decent drinking water, except for it being quite hard.
Never had issues without filtering it first either.

Is it just me, or is something really off with the blue colouring?
Anyone else having crappy growth on coloured plates?
I thought at first only some red ones were anti-fungal.

All of my blue plates look similar to those above.
Can't be compared to those without colouring and BRF in addition to ELME/LME.
