Keeping this old thread alive lol

So I had to start over but good news is I got a new 2x4 ft FFU so things are going much better in that department,

I’ve been working on a lot of projects lately but I decided to start over with the morels so the other day I formulated a new agar mixture for morels specifically based on several scientific articles and adding my own twist

Anyways I dropped some penn state rufobrunnea sclerotia pieces on the new agar and I also put a random black morel a friend of mine donated from an LC on the new agar mix

Here’s some pics of the explosive growth after 2 days

So I have some beautiful, aggressive, clean black morel plates cloned from the wild and I have no idea where to go from here to get fruits. Any good ideas?

Just for the sake of archive, those private game plans could be beneficial to the collective, even if failed.

It wasn’t anything that hasn’t already been talked about here 100 times over lol I was just trying to fast track and get him caught up without cluttering the thread it was a summery of what’s in this thread

Also your plates look nice good work, what was your agar recipe

Most of them potato flakes boiled strained, and some karo... maybe a little  powdered dextrose I had left from whiskey making few years ago.

Possible the was some nutritional yeast in some of the.

I've got other plates that are so dark they don't photograph well.

Hello, I wanted to know if you have a technique to manage the formation of sclerotia (pseudomona reserve) on morels?
And if you have successin fruiting them from one year to the next.

Sclerotia is not a pseudomoa reserve it is a nutrient reserve comprised of lipids

Only natural morels that form symbiotic relationships farm bacteria but the landscape morels which are generally associated with the elata clade or rufobrunnea clade don’t need bacteria so that would mostly apply to the esculenta and possibly burn site morels

Elata clades are heterothallic so they need both a and b mating types

Rufobrunnea is homothallic and therefore doesn’t need both mating types because it is self fertilized

The key to larger stones is nutrients and age of the culture, morels are Ascomycete like cordyceps so they become senescent the more transfers you make which reduces their metabolic capacity to store nutrients, this is why morels cultured from spores are much heavier stone producers

This is also why they shut down the commercial morel farms in America because of strain degradation and parasitic molds on the fruit bodies

Ok,thanks for the answer,
I believed that the bacteria which go dormant were ingested and stored in sclerotia to serve as a reserve. So I did not understand the mechanism.
thank you for your explanations.

No problem

I do believe however, that soil bacteria like pseudomonas are possibly beneficial in the casing layer like with Agaricus species to trigger pins but that’s just a theory of mine, it would make sense why those who have been successful don’t seem to sterilize their casing/soil

Now i don't know ifthose informatiin are exact, but
during a podcast a producer explained that the failure in year two of production was due to the fact that the mycelium had absorbed most of the bacteria and therefore for production over several years it was necessary to get around this problem.
Links i hope there is a translation it is in french:

Ps:these sublect is very intersting and as homegrower amateur it would be pleasent to griw a few morels.

all 3 of the (likely Morchella importuna) cultures I started have made some sclerotia on the side of the jar of agar they're in, less sclerotia are on the actual agar, but they're there too.

So I recently made an interesting observation and it’s funny how it actually correlates to the topic we were discussing with soil bacteria interactions, granted this could just be a 1 off case but it’s still worth noting

So a few months back I made 100lbs of oats and I inoculated 4 10lb bags with importuna I acquired through a trade but the vendor they received it from was sporeworks, it was unopened so assuming it was still sterile I banged it straight into the bags

1 of those 4 bags contaminated with some type of bacteria which for lack of better evidence I’m going to assume is a bacillus species because the filter patch smelled of sour rot, so I set it to the side but never got around to trashing it, well after a few weeks away, I came back to find that it had the most explosive morel mycelia growth of all 4 bags I mean it even started making a condensed snow looking mat, but it was definitely morel mycelium

As a test I took another bag that was clean and I did an aggressive break and shake that always results in the filters failing in my experience and allowing that bacteria to enter, this resulted in faster growth and denser
Colonization

I just did the same to the other 2 bags last night so we shall see what happens with those as well, and one of them even broke the heat seal at the top so i know for certain it will get contaminated but still worth observing the effects

Now previously I’ve had this happen with rufobrunnea and they do not tolerate it at all, they seem to immediately stall out so this suggests it is at least clade specific or even species specific but it surprised me because importuna are a landscape species so they are saprotrophic and grow well in sterile culture

This observation doesn’t necessarily mean they are feeding on the bacteria it could be more of a stress response to gather as much nutrients as possible to outcompete? Not really sure but it’s interesting

Something worth mentioning though, is that none of the bags from either species has made stones contaminated or not, there’s zero sclerotia present, my bags are plain oats so it’s probably too nutritious

Now my plan for these bags will be to run more indoor trials but some of them will get loaded into a grass seed spreader and I’m going to inoculate my entire yard with 10-20lbs of colonized grain

I have 2 rufobrunnea and an esculenta culture.
I made a couple of 6qt shoeboxes by layering 1qt oat spawn on the bottom then just barely covering that with pasteurized sand, then about 2" or so of 90% Peat/ 10% wood charcoal + oyster shell grit/gypsum/sand. The rufo's shot straight thru it and colonized the top layer completely with a fine cobweb like myc. They're starting to make orange sclerotia in the grain layer as well as the top layer.
The esculenta isn't going fast, it's just starting to colonize the top layer, and it looks like typical myc. I think they would have done better with a traditional mixup at spawn.
I just got 2 importuna cultures,  so I'll be trying something similar with them soon as a baseline.

A possible suggestion, you might want to take off some of that peat casing according to the patent and other articles it seems they need to be shallow just at or under the surface

But that’s just a suggestion, not sure if it’s “critical”

I made 2 bags each of:
1. M. rufobrunnea (strain #1)
2. M. rufobrunnea (strain #2)
3. M. importuna (Sporeworks)
4. M. importuna (Mycelium Emporium)
5. M. esculenta (MushroomManMycology)

Sub mix is 50/50 woodchips & HWFP / spent cube subs(mostly coir) with some gypsum, sand, and perlite. Most of the bags look like they're producing sclerotia.

So as unusual as it sounds I think I want to try something unconventional,

Bag cultivation of morels, like actual fruit them inside the bags

I’ve seen it done once I believe it was the Israel MIGAL institute with professor Segula masaphy using a rufobrunnea culture

But I think it might be worth a shot even though it’s not really a well known or even considered option

I have some nice sclerotia grain spawn to throw at it
