Ok, that gives me a good reference - I'm starting off with 30 glass plates and as I get extra $ to invest in more, I'm going to try to get up to pouring around that, maybe a little more.

I use 75mm plates, I usually get 21 to 28 plates per 500ml bottle depending on how thick I want them.

Good to know, thanks for sharing. I have 100mm plates so I think if my math is correct, I should be able to pour around 15-20 plates per 500ml bottle.  I'm ordering 2 more bottles so I'll have enough and won't have to stress

Do consider what others said. 20-25ml per plate is pretty thin pour. It's preference really but who knows you might prefer thicker plates too

Pour em thick! Thin plates are a pain.

I use thicker plates if I think I might be storing a mycleium plate in the fridge long term.

I also pour it thick if I'm hunting for plins to clone.

Deeper plate (and possibly a little more water than usual) will help keep it from drying out in the fridge, or while you wait for plins.

I think there are better ways (than slants) to store long term, here are a few links I'm aware of.

Easy LI for Preservation and Propagation
Cryogenic storage of P. cubensis mycelium, or can I freeze my cultures?

And when we're on the topic of slant's, has anybody ever made small, micro slants using these? I really don't have much space in my fridge ... I'm wondering really, what are the benefits of the bigger centrifuge tubes vs agar plates anyway, except that centrifuge tubes close them self's up better and thus maybe don't let as much moisture through (all my agar plates in the fridge end up having water in them, in spite of having them locked up in sealed Tupperware).

I’ve used these to send wedges of agar with culture, and have received these from others. I have a bunch that are almost a year old with usable cultures still inside. I’ve stopped using slants. There are other easier ways to store cultures.

:whathesaid:

They dont even need to be in the fridge.
I've had better luck with older wedges in these ,Than I've had with newer cultures sent in Dh20.

Maybe it’s just me but I didn’t quite get if your main problem is getting contamination even with plates you haven’t even used yet, or problems after inoculation, transfers? This probably influences the discussion quite a bit.

I've gotten a few plates with what is clearly edge contamination when using the pour methods, I haven't been sealing after pour/before use but I have stacked and covered them in the SAB. I've also gotten the occasional random splotch which I attribute to my SAB/hand technique when pouring(all before use).  I didn't consider these a huge problem - just another factor that lowered my yield of good, usable plates from a pour session or technique.  With the Primo Plates tek the contam issues have all gone away but condensation is not satisfactory (for me). Were all 60 of your SAB pour plates usable?

I'll emphasize that I'm quite new to this. So I can only answer from my limited experience.

Out of the 60 plates I've poured, 1 was unusable, because it had some kind of mold growing on the edge, after about a week. 4 plates from the first batch were ditched even before pouring, because it was my first time working in a SAB and I fumbled getting these out of the bag, so it seemed safer to just ditch these. Everything else has stayed usable.

I too do not wrap my plates after pouring, I just slide the package that they come in back over them and store them in cupboard (feels a bit finicky, but haven't messed it up yet).

I get quite a bit condensation even with poured plates, even if I let the agar cool down to a bit under 50° C. Nothing that I myself couldn't see through, but complicates taking pretty pictures of them. Maybe I should let them cool a bit in a SAB before sliding the package back on, or maybe it's just my environment. Doesn't bother me too much.

Based on your description I personally have a feeling that something is going on with your SAB technique when pouring if you are getting results that do not seem to satisfy you. Maybe you could describe your pouring process a bit and someone more knowledgeable can point out something. For example I split the 20 petris to two piles, so that I can manage pouring them by lifting the lid of the bottom one, along with the whole stack and work my way up. This was probably described somewhere.

Yeah I cut my plates in  stacks of 5 as to not knock em over.

If you're not using a media bottle with a drip ring that may be worth investing in ... or I've seen ppl use liquor spouts which look pretty nice for pouring.

Only open the lid just enough (of plate) to pour and close promptly.  And elevated of the table...

Good point about the media bottle, got it right from the start. Would probably just flip the table out of frustration without it.

I have a couple media bottles I use- when I poured plates in the SAB, I'd split 500ml batch between the two bottles so it wasn't too full for those first plates.

My latest try I used the no pour Primo tek, then when I go to use the poured plates, I pull a fresh sleeve of sterile plates out and carefully swap out the "condensated" top  with a clear sterile top.  These all look great so far - might be my method of choice going forward.

Definitely Wasted my time on this go around... So I think, I kinda suspected my liquid culture was bad but.... I went for a test run on some agar. Here's the outcome so far on a bad plate.

Will an LC test plate typically come out looking this shitty when it's bad LC?

Lmaooo I'm jc bc I had test plates that I'm 90% were good and then most everything shit the bed anyway. Im jw if bad LC will look obviously messed up like yours when testing on agar. I feel like Lew told me that it's not a guarantee, but my memory is dog shit.

I wrote this a while back, and I post here to anyone wondering about LC not just you:

Liquid Culture. Don't do it until you master clean agar plates.

LC can be a great way to grow a lot of shrooms fast, but it's also a great way to grow a lot of mold and bacteria fast.

I don't see the point of doing LC early on, or when still having any contam issues.

LC is unneeded unless you're running a commercial grow and need to grow kilos. If not, why not just toss agar slices into grain jars, and shake at 30 percent? You're much more likely early on to get clean grows.

It's true that you can't always tell if it's clean in the liquid, but if someone must do LC, I'd recommend start using nothing but 4 grams dextrose in 100 ml distilled water, PCed for 25 min, cool to room temp before nocing.

It's as clear as water, which gives a better chance of seeing contam.

Good LC basically looks like sperm in water. (still should be tested on agar)
