How about the LC? Am I thinking about it right that all of the liquid in the syringe is an inoculant or is it only the part w visible myc floating?

Your thought process is correct.  It’s recommended to vigorously shake your syringe prior to use, but a good LC should have myc throughout, even if not visible to the naked eye.

Sometimes, they just don’t take on the agar though.  Good luck.

Edit to add make sure if you are flaming your needle prior to innoc (as you should be) make sure to squirt out the LC that was in the needle and use fresh LC before you put it to agar.  If you go straight from flame to agar you are using dead LC.

AA+ plates, looking good, feeling good

I want to put them to grain in about a month, can I just bag them and pop them in the fridge?

cheers all!

Really? Will do a transfer to new plate then, thanks. I don't know if it matter but it's riptide OG.

I keep plates in my fridge for a year or more and they're fine. Just don't freeze them. And expect a little condensation - it won't hurt anything.

Good points. I'll add that I store live plates in the fridge in a plastic zip lock bag, and store them upside down, so the condensation is in the top of the plate instead of on the mycelium.

Unrelated pic tax.

looks like tomentose mycelium to me. if it's not you'll know very soon lol. if you can get your camera to focus on the myc instead of the cover of the petri dish it'll make things easier.

No they will not. The clone will outgrow MS long before the spores even have a chance to germinate. clones start growth immediately. Spores do not. You take a transfer from that outside edge and have absolutely zero chance of grabbing multispore. I do it all the time.

I do the same, if anything I cut of the base of the plin to remove mycelium that is not part of the fruit. This discussion made me wonder though - a mono that germinate from a spore could easily fuse with the existing mycelium, and its DNA could propagate to the leading edge.

Yeah that was my initial thought too. Well, time will tell, needed to spend some time in the SAB today anyway, and based on previous answer I did 2 transfers to new plates. Thanks

I didn't even think to ask that question, thanks for pointing that out. It's T0(zip wedge to agar) so it's not going to be clean, but I don't want to isolate mold...... Again

haha, I feel you. I'm still trying to learn to clean stuff up with ah shall we say mixed results. so make some transfers but also maybe look into pucks?

edit: thought you were distinguishing between mold and bacterial myc, like maybe would abandon the former, try to clean the latter

For sure, I've been at this hobby for 2 months and all I have done is agar, the thought being that if I spend extended time with each phase of growing I might make fewer mistakes, and maybe learn what to look for in each phase. It feels kind of cruel, but I have been using oyster to learn.

Have probably done about 120 plates and with the exception of the 20 first I did from 2 plates I got from someone else(which I was told by others on here where pretty dirty from the get go) I think I am doing okay and hope to move on to grains soon.

I can't get brown rice flour where I live, but I can get brown rice and have found som good containers for brf pucks, but I need to get something that could crush my rice in to flour.

I feel pretty good about my Nat plates, and was thinking I could do 1 more transfer before I go to grain, what do you think?

I like that strategy, amazing. I tend to go off in all directions at once (and it often bites me in the ass) I'm guessing you live far north? Grinding flour seems like a pain in the ass, but on the other hand you need so little per puck it might be worth it. I should probably wait til my eye is better before commenting on people's agar plates. Hard to keep quiet sometimes, especially when someone's question is left hanging awhile.

That said, those nat plates look good!

Yeah I did the opposite when I started homebrewing 7 years ago, and shit, it looks like I finally learned from one of my mistakes

If I remember correctly I could grind the rice with the same tool as I will need to make mushroom powder down the line, what are people using for that?

And I'm glad you answered even if it turns out to be mold all over that plate, because I already learned something from your answers and that is to be more specific in my question if I want detailed answers :toast:

Coffee grinder works really well for both for small batches. For bulk mush I have used a ninja blender most often.
