They are 3 cultures that germinated from the same mushroom's spore deposit in that mason jar.

2 are from the same 'petri dish' germ plate, but deliberately taken from different sides.  2:00 & 11:00 ->

1 is from anoter germ plate entirely.  Scalpel was flame sterilized in between transfers.

so they are all new cultures 'strains?' from the same mushroom, but from different spores.

Ah yes then i suspect the one that hasn’t darkened yet is just a slower isolate

I think the proper terminology would be phenotype, variant, isolate or subculture but the strain would be “importuna” according to Alan Rockefeller

If you’re making an LC I would suggest potato dextrose broth or malt yeast peptone broth although karo LC works just as well so maybe not as important at that stage and karo does offer a better clarity overall

I would just mix the sawdust and grain evenly but if you want to follow the patents they actually layer their bags

Top 50% is potting soil
Bottom is 50% wheat grain

And they inoculate the potting soil side to force it to grow towards the grain but translocate the nutrients back to the potting soil layer
Later after a month of colonization the separate the 2 layers and spawn the potting soil layer into a sand soil mixture

I was just thinking about grain soak water, I'm about to soak some more grain.  I have more than I need.

I didn't want to buy cheap sugars to make an LC...

Thanks for planting that seed of grain soak liquid culture.

Edit-  I just read hamloaf's HLC thing, and he does 1 part GSW:4 Parts Water...

I'm going to try that

Here are some clearer pics from today. I inoculated these jars 2 weeks ago(maybe 15 days), but almost seems that would be too fast to be like the yellowing oat jar pics if it is healthy sclerotia after seeing some from what I read somewhere. The the sclerotia of these ones seem to really love oats looking at where the sclerotia are forming in any jar. I am going to prepare a few jars of LC and more oats(but G2G this time) in the next few days to try a bunch of random stuff indoors, my backyard, and to get them growing in a few people's yards. I didn't take any of the jars with normal lids or a small layer of grain under the sawdust mixture. They seem ok I think, just a lot slower and no sclerotia yet.

The jars that are completely yellow/orange are healthy af good work, yes it’s totally normal to see explosive growth with morels they can completely colonize a substrate in 2 weeks

1:4 GSW:water for LC's, to inoculate soon.  Some are 1:2 to experiment.

LC's in quart jars... bad idea?

I would have recommended a modified lid, the size of the jar isn’t as important if the lid is modified for your needs, I personally prefer a syphon type lid with a self healing port and a syringe filter

For shits and giggles I decided to play with some unconventional ideas, I took the “raft” of floating morel sclerotia out of the LC and washed it with tap water then loaded it in the blender with more clean tap water and made a slurry

Then I grabbed a turkey baster and dropped the liquid sclerotia onto 2 lbs of pasteurized composted straw/manure just to see what happens

I still have some slurry left over so tomorrow I will drop it on some sandy soil mix and then place some ENB’s on the inoculated soil surface

The shoe boxe totes with white lids are Clitocybe nuda and the gray lids are morchella importuna liquid sclerotia on compost

Been almost a month without updates, I’ve been busy with other species and financial instability,

(Also the morel slurry on straw compost i mentioned earlier failed miserably)

I got some clean LC’s made from the penn state morchella rufobrunea cultures so i know they do much better on a high nitrogen medium so im doing some testing on different nitrogen supplemented oat spawn,

The first one is yeast/peptone  (y/p)

Then soybean meal (S/B)

And finally coffee (c) which I’ve used in the past

All the jars have gypsum

In the pics you will see blewits are also being tested on this particular batch but just ignore that

Your absolute right man and I do apologize for being so rash it wasn’t right for me to call you an asshole …

…. However I do want to say you really are wrong about that being bacterial and I stand behind that 100%

Also in my honest opinion, it is morally wrong to tell someone their work is nothing more then “ripe wet bacteria” and to be so brazenly wrong about it, which is usually followed by “just trash it” especially without knowing the way this species mycelium behaves so he would have tossed out perfect jars because of your lack of insight.

I hope that you can accept my apology and we can move forward but at the same time I also hope you understand why it is upsetting to see you and others perpetuate this all or nothing perfectionist mentality, and it’s not just you it’s the main stream culture that’s really pushing this but so many people say “contam” and it’s like you just put a death sentence on them when it’s just bad identification because of lack of information

Hey
I finally acheive to clean some plates. I had not the time to make potatoe dextrose so only LME. I understand that is not the best to see big sclerotia but still, I thinks that those black point are indeed?
2 first are from the same plate, one darken the other not. In fact I can see way more dark point in the dark one so make sense that the color change could be metabolite reaction when morel create those.

And the 3rd is from another strain

I will tranfer on some grain next week, is there better grain for morrel?
I only have millet for now but I have to re stock so I can take anything is great for them. And I will do only grain and also grain+sawdust

Whole wheat berries or rye is what most papers describe using

The black spots look like clumps of undisolved agar

Sclerotia will always form on the surface and are orange or brown rarely black

If the black spots are on the tips of mycelium it’s possible it is mucor mold

But morel without nitrogen in lmea can present similar just takes some training of the eyes to learn the difference

Here is an example of morel sclerotia on agar. This is esculenta.

I'd say your black specs are sclerotia. I've seen them fully pepper plates before. I've also seen big monsters form in plates where only 3 or 4 will fit in a 90mm plate. Interesting how contrasted to the mycelium they are and how they are forming underneath the agar in your plate.

Re: morel cultivation thread [Re: Nichrome]
    #28834132 -

I agree with nichrome

It’s possible that is sclerotia but I’ve never heard of or seen it form under the agar unless it’s just the trick of the camera, A good way to know for sure is if the agar and mycelium begin to darken  like in your first pic
And the one nichromes shared

The one that’s entirely white with black dots does make me wonder though because usually browning of the mycelium and agar happens at the same time as the stones appear

Once it goes to grain it should be evident what you have

Re: morel cultivation thread [Re: 7Suns]

Updates on my rufobrunnea test jars, I want to start by apologizing for my inconsistent parameters because I’m working with sub prime equipment in a sub prime environment, during a financially trying time in my career, so in my efforts I actually overheated my grown out LC masters used in my test jars of grain, ac went down and they hit temps of 105F for about an hour or 2 approaching threshold for thermal death, without taking this into consideration I still inoculated my test jars and for the past 2 weeks nothing has happened until today when i checked them again because I assumed they were toast so i was ready to toss them and start over

Well they finally showed signs of life but only in the soybean/oat jars

Also I imagine there’s a lag between switching from simple sugars like karo or malt back to starch like grains, I have witnessed this several times now with this species on agar going between lmea and pdya

I am 100% certain that this is rufobrunnea mycelium based on the way it has white fan tips but angel hair thin mycelium and then the mycelium back towards the center is visibly yellow and oily looking and almost becomes translucent

I witnessed the same morphology with this culture on PDYA plates but on malt it never develops the white fans or stones

So now that I have an idea of what too feed them I will try to mass produce some 10Lb bags, I’m just glad to see some good mycelium plus now I can use these test jars for G2G
