Thanks all work was done in a still air box trying to be as sterile as possible. But yeah the condensation was pretty heavy on the PE Clone. Here's an up close pic and another with a light behind it. To my rookie eyes they look pretty darn good as well.

40 plates poured, 4 different recipes (10 of each).

Recipe 1 (bottom left)

200ml distilled water
3.0g agar
3.0g light malt extract

Recipe 2 (top left)

200ml distilled water
3.0g agar
3.0g casein hydrolysate

Recipe 3 (bottom right)

200ml distilled water
3.0g agar
3.0g light malt extract
0.1g nutritional yeast

Recipe 4 (top right)

200ml distilled water
3.0g agar
3.0g casein hydrolysate
0.1g nutritional yeast

It's an experiment. I have 2 clones, 6 various transfers and 2 germinations to do. For each inoculation I will repeat 4 times across each of the different recipes. This way I will be able to compare like for like for each of the recipes in different situations.

I poured them a little hotter than I usually do due to time constraints so there is probably a fair amount of condensation. I'll leave them in front of the flow hood until tomorrow and do the inoculations at about +20 hours.

Initial observations are that light malt extract is more orange where as the casein hydrolysate is very pale. Nutritional yeast makes the recipe cloudy. Casein hydrolysate is much nicer to work with than malt extract.

Why did you try the casein? It's a protein so your swapped a sugar for nitrogen... its not a different kind its an entirely alternative form of nutrition

These are transfers I took from agar I colonized with grains and a couple pins. The pins all look like mold. I had trouble so I'm not surprised ... anybody have good advice or a link to a page that gives good advice about cloning pins. It was harder than I thought. I couldn't get a piece to stick to my razor so I kept having to cut it and stab it over and over. Not so good... anyways the grain transfers look good to me what do you guys think? Should I do another transfer on the good ones or are they ready for grain. I can take individual pics if anyone sees anything they think I should address. Thanks in advance. The top one and bottom two pics are the good ones and all the others are the pins that sucked.

Is it normal for the mycelium to be aerial near the agar piece, or is that contamination or what. The petri dishes look funny compared to my holy Grail no pours, but i also cooled my blade all over in them, hence why it looks broken in some areas. What do y'all think? How should I proceed?

I don’t have any particular expectations, in this case I am just curious to see how the fungi behaves in different situations. I’ll repeat the same clones, transfers and germinations on all four recipes and observe the results.

Not sticking up like the Petrie dishes and they don't look funny even though there are huge razor gauges from cooling a red hot blade... What gives? Any advice would be appreciated. Do I need to keep transferring or are they clean?

The aerial myc near the transfer piece is very common ime. Nothing to worry about. I believe that it is just the myc stretching out and "looking for" a hospitable place to grow before it finds the agar surface.

Not sure why you don't see in on the holy grails. It just may be harder to see due to less clarity?

It looks like you are cooling your blade near the center of the plate where the transfer goes. I recommend cooling your blade near the outer edge so it doesn't interfere with the growth. That just makes it harder to determine if you have nice symmetrical growth.

You can certainly do it that way as well. I take tiny transfers and find that the hot scalpel melts the agar making it hard to see my target piece. Like  tiny little 1mm pyramids.


If I cool it in agar, I usually do it on the outer edge of the transfer plate rather than the receiving plate, but that may not be best practice since sending plate has a live culture and ideally the receiving plate is completely sterile until inoculated.

Sent some tat v wombat swabs to new agar recipe (6gs LME/8gs Agar/400 ml distilled) only did one Z shaped streak to not have such messy plates. Most of my other plates look really moldy so I tossed most of my germ plates and only kept one ghost plate in hopes of pins… I haven’t gave up just yet folks. I’ll give it my all till I run out of spores :facepalm:
