Did you use fresh spores for these? I swabbed some plates this weekend and I didn’t see any black streaks and last time this happened they didn’t grow. So just wondering if all the spores I have are old or what gives.



They are definitely old... The Golden Mammoth is from 1/20 and the rest are some I won in a giveaway from a dope member here in 2021 that I haven't gotten around to yet. So I guess we'll see :dancer:

The ones you see with black streaks were from swabs caked with spores. If I had swabbed that APEU plate instead of left the swab in you wouldn't see anything on the plate. And yeah sometimes they just don't come back to life. I've heard "wetter" agar (less agar in your recipe) is helpful, and I feel like I've seen teks on how to best work with old spores but I can't remember now

Gotcha… ya I think is pretty “wet” compared to my last recipe so we will see it’s only been like 3 days on these plates but the one with visible spores on it is like 12 days old so I might just have dead spores. No wonder I’ve only been able to grow mold from swabs.

Spore syringes are perfect for agar.
Now you transfer the clean growth and don't use the syringe again.

Any recommendations for a source of pre-made agar dishes? continuing to make my own may cause discord in my home, so I'd like to have a backup source for when I run out and can't make fresh ones.

First of all, I know these pictures are blurry as fuck, and I'm sorry.  I swear I'm going to have to buy a camera just for these macro shots.

Anyway, these are nats.  They were looking pretty solid for a while, but as it grew toward the edges more, the growth started to change, especially on those first two.  I don't know if it's just nats being nats or they're still dirtier than I thought.  These are T3.  Any thoughts?

Looks free from visible bacterial or yeast contamination to me, mold is more difficult to say. How long after inoculation were these photos taken? And how did you inoculate them?

The syringe was squirted onto the agar liberally? The reason I ask is the cultures growing from edges of the plate are more suspect in terms of environmental contamination.

They look ok germination plates to me. I'd transfer some of the cultures until you get uniform growth and also keep the old plates for observation.

All other things being equal, if you have 4 transfer plates from the same donor plate and 1 or 2 have satellite mold colonies on the edge, is it best just to ditch those plates and transfer from the ones without satellites, or if the myc in the center looks equal to the other plates would you transfer from those as well?
The answer seems to be common sense but I would like to hear from others' experience.

I rather just run a torch for like 5 seconds and cut a triangle than spending all the other time to make a perfect circle agar wedge personally. To each their own.

If I make a mushroom shaped punch, or...any other specific object shape -- and have a healthy culture going, will the plate grow out like the image?

The worst is when you cut a perfect little wedge but it won't come off the scalpel onto the new plate so you need to stab and twist the fucker until it has no choice but to enjoy it's new home. I'm sure that's terrible sterile procedure but not sure how else to approach it.



Picture tax. Are the little darker spots in the mycelium contaminants? They appear as the Myc grows out so I don't think it's little LME nuggets.

This is my last plate with food colouring too. Going clear from here on out.
