you can kinda get away with unwrapped plates but success probably depends on environment a bit & you gotta be so delicate when handling. a layer of parafilm/cling wrap is pretty cheap insurance from user error ime lol

I don't think I could give up my Pyrex petri dishes. They're a staple in scientific and industrial applications anyway, so I don't see why I shouldn't be able to make them work.

I'm also trying to minimize my single-use plastics budget.

My blanks often stay unwrapped in my closed up SAB. I lose one now and again, but I also reuse disposable plates and don't always turn off the AC when I pour. So I have multiple sins to pay for. 😂

the pyrex ones are super nice, would definitely be silly not to use them if you already have them. if you’re having trouble with pouring em, try using them no pours. way easier & more consistent than pouring them imo

I tried a couple of plastic options but I do like glass petris. I also need relatively few going at any time.

Thought, I can't beat condensation with any kind of dish, pour or no-pour.

100%, I switched to plates from no pours not long ago and forgot to get grafting tape or parafilm when I bought my dishes and just sent it anyways. It’s been working out so far so been lazy about it but it’s definitely a pain trying to handle them and wouldn’t recommend for in-use plates.. I’ve got a couple I don’t want to risk it with in ziploc bags for the mean time, it works decent but clunky

for sure dude i feel you, when i first got my plates i didn't realize wrapping petris was even a thing :lol: i had tried ziplocks but they molded p quick on me. i think that was my fault for being sloppy tho bc i know they work for other people. yeah too much can go wrong when unwrapped & colonizing, at least if you lose a blank dish it's not too bad. that's how sirP does pp5 petris i'm pretty sure. the glass are just kinda slippery & loose without a wrap

Dude that makes no fucking sense. Have you actually managed to grow cube myc on a plate with that recipe (which I assume is 2/2 or whatever close enough to that) ? If yes, then you should be able to grow everything else in there as well.

Are you using tap water or DI? Very long shot but you might have some ridiculous antifungal/antibacterial additives in there that survive the sterilization process somehow.

Basically the gist of it is like somewhere about a year ago my agar started giving me troubles, I was also going through life stuff so the troubles stayed relatively un-pinned down and I also had slowed down on mush cult, last little bit now I've been troubleshooting and trying to figure out 1.hot break and 2. Why my agar fucking sucks

My last batch that was the 2%/2%, 2%/1%, and 2%/.2%, had absolutely zip show up on the open air and spit plates. So now I'm here, telephone agar and Briess Maltoferm 1001, 1.375% agar 2% nutes, sterilized ~according to mycolorados chart (36 min for 400ml).

It does appear to have fixed most of the hot break issue, but now we wait to see if life is still allergic to the plates.

Its been a bit frustrating and I think I've scrapped 80-100 plates in the last 3 weeks

Oh also the test plates that grew zip where distilled water, these new ones are tap

I don't know wtf you guy's are doing to manage this but there's no chemical or physical explanation for "dead agar". Agar isn't available to the mycelium as an energy source due to the carbohydrate structure of the molecule, so that's not it.

Even if it was your malt, most bacteria and target mycelium will grow on water agar void of nutrients.

Also, stop using spit as bacterial inoculum. It's not going to be reliable:facepalm3:

Yeah thats what logic would seemingly dictate but alas, 20 fucking water agar plates no growth whatsoever. :shrug: something is obviously wrong, when I hold a flashlight in this room at night there is a whole cone of dust and particulate, I know spit isn't a reliable inoculum, I dont have plates of sample bacteria stored away for this, by any means a plate opened in open dirty ass air and then spit on like a kinky bitch should not show ZERO growth, Briess malt, Telephone agar, distilled water, following your recipe.

Its annoying as fuck. I've read before that some folks had success with problematic agar by lowering the hardness, so thats what I'm trying. If this shit doesn't wanna grow anything I'll buy new agar, as thats the only variable left ffs

@Hobbit, I did with this latest batch, I followed mycolorados chart which includes the vent and coming to pressure times, so overall worked out to a decently shorter cycle

I am 100% confident that your cycle time is irrelevant. If you could produce a gelling agent impervious to microbial activity, you'll probably win the Nobel peize for your engineering achievement.

The only folks with this dead plate problem are noobs. Not one TC has ever experienced this issue, so far as I'm aware.

I  challenge you to go outside and find some mold, place it on this dead agar, report back.

I do think food coloring was fucking me over for a while and I've sworn it off until I'm an agar master, I think the water plates having a couple drops of blue could've been their curse but idk, gotta troubleshoot more

Multiple open air, dirty finger dabbed spit on it, over a week and nothing.  Telephone agar and briess CBW golden light at 2% and 2%, distilled water, sterilized following stipes procedure.

It's not your agar. The only time you would have agar problems is with stubborn spores that need a bit more hydration. You lower the agar concentration but keep the h20 constant, they get the extra water and the baby magic happens.

Fuck it, leave a plate open in your bathroom. If nothing grows after you flush, you got problems.

Stipe's spit is probably sterile because of all the booze. I know because my breath is like a distillery. Get a Komodo dragon pet, then have them lick your plates. If you can't get some bacteria going then Psych's right, you have a forcefield.

It is pretty weird though. We put so much effort into making sure not one speck of mold or bacteria touches our plates or they contaminate. And they can't get their plates to contaminate if they try? Weird.
