Get a camera app that has a focus lock feature. OpenCamera on Android. Turn that on, tap to focus once, then adjust your distance while zoomed until the myc is in focus instead of the plate. Then zoom back out while in that position. Easier with a tripod.

#1 & #6 look the best to me, most uniform growth. But the rhizo in #5 on the right is probably worth grabbing too.

:whathesaid:
Adobe Lightroom is what I recommend for iphone. It’s free and allows you to fully control the camera and export close to raw instead of fully compressed straight to the camera roll. It’s what I used for the pic in my signature.

Just so I don't fuck it up, from stalking you guys on this thread am I undestandig correctly that I'm supposed to cut at the edge, again a small square or triangle?

How many transfers should I make from one plate? Last time I did the T1 Kat suggested making 6 plates from the healthiest looking germ plate spots, straight out the fluffiest middle part.
Is it enogh to do like 6 T2 plates from 1 or 2 of these T1 plates?

I will try the app thing next time I post.
Thank you for the advice.

i like doing a tic tac toe kinda cut that forms tiny diamond shaped transfers.

Xd, thanks, maybe i'm too pedantic regarding all the details, I was more interested in asking about taking from the edge of growth or is it not important if it's the edge or middle or wherever?

These are my first ever transfer plates from first ever germ plates, please bare with me.

Do I have to let my plates fully colonize before going A2G or can I treat it like a transfer? In other words, once I get a quarter sized sample take a wedge from the leading edge to transfer to a grain jar? My thought process is the less agar I use to transfer, the less chance I'm transferring contamination. Obviously "it's faster if you use a whole plate", but from my own experience haste is the hobgoblin in all aspects of this hobby.

The reason to let it grow out some is to see how it looks so you can be sure it's clean. If the agar was contaminated you see a satellite growth on it and would know. Sure transferring a smaller piece means that if something was there you could possibly avoid it. But that's not really the view to have to goto grain, you goto grain when the culture definitely visibly looks clean.

Hi everyone,

what do i do with T1 plates after i did a T2 transfer?

I resealed them.
Can i keep these for reserve in case T2 get contam and then do another T2 transfer for example?

Should I put them in the fridge? In a couple of days the myc should reach the edge of the plate.

Now i'm wondering how to best use the T1 plates, so that i'm not throwing away something that is still potentially useful.

Peace out.

Yep you can go back to the T1 plate and try again if T2 plate doesn’t look great. The fridge will slow down the growth so that is definitely a good option if it’s almost to the edge. You can keep the T1 plate at room temp and it’s possible that the plate will pin then you can transfer that pin to a new plate for clean growth and stronger genetic chance. Other than that no need to throw the T1 plate out just keep an eye on it for a month and see what happens

Thank you sir.
So transfering a pin would mean taking a piece of the "fruiting body" (I guess) and putting it to agar, right?
Or am I mixing up something here. I read in Mckenna's book about taking a piece of flesh from a fresh cap of a mushroom and transfering it to agar as means of getting only one specific "strain" of dikaryotic mycelium.

Is what you are talking about similar/same?

When you have a pinning agar plate, you can transfer that pin straight to another agar dish or just straight to grain. In a SAB, I just grab the pin with tweezers and toss it in a grain jar for use in a quick clone shoebox.

A pin that grows on a plate. If you want to increase odds of finding one, keep your germ plates around. They fruit easy.

The idea is that a mushroom that will fruit in those conditions is strong and may do well when put to bulk. People clone or send plate pins all the time.
