I always let my plates grow out to the edge of the agar, unless a contamination rears its ugly head. (In those cases, I always take my transfers as far away from the contamination site as possible, and as soon as possible.)

If my germ plate never shows any contamination, I send the germ plate to grain without ever transferring off it (unless I need to make more plates to knock up a specific number of jars - I usually do one plate per jar planned).

Yeah I wait a little to see if it goes rizo. if I never see rizo then I’ll take a piece of leading edge. I had a germ plate that didn’t go ropey till it almost hit the edge of the plate.

Thanks for the advice. What are the visual markers for the certainty of contamination, the uneven & sporadic growth coupled with stalling? I'll be taking that transfer today, nice one.

I started having problems with condensation with my no pours after I switched to a different PC. I used to let them sit in the PC and cool overnight. I think what happens in this scenario is the glass and the water cool at different rates, causing condensation on the glass in the 100% humidity enviornment.

Try this. Either raise the jars or make the water level a bit more shallow so it doesn't make contact with the bottom of the jars. When the PC cycle is done, let the pressure release naturally. Then take the jars out while still hot. I use unmodified lids and leave them a bit loose while cooling and after inoculation. This solved my condensation woes.

Many say the condensation isn't an issue, but you have to think that if there's any contamination in there the water's going to spread it everywhere and contaminate everything as soon as a transfer is made. In other words you won't be able to outrun the contam. Also made germination impossible for me.

i dunno man

my lc test usually look like clean MS germ plates. that looks like bacterial syringe water. maybe its just the pic:shrug:

It's from this Z-strain plate which was originally looking great on the dish, really nice even rhizo growth.. but it keeps coming up bacterial or tomentose whenever I transfer it :/ This is from a Plate Pin also, idk if that'd make any difference.

Probably down to my technique I suppose.

Onwards & upwards eh.

I was checking back on a plate that I took some t1s from about 1 week ago and saw some super ropey rhizo growth that wasn't there when I took my original transfers, so it got me thinking



1) Inoculate plate with spores.
2) At 50% colonization, take transfers of fastest growth.
3) If transfer plate looks clean, allow to fully colonize and pin.
4) Transfer pins straight to grain.

I use it only when I’m cleaning up a culture. It really does make a difference versus just straight water agar to save cultures from bacteria.

Never send Rit agar cultures directly to grain, though. You don’t want to consume the ink. I always return my cultures to nutrient agar that is safe to send to grain or LC.

Here’s my recipe; it’s enough for 15 plates:
500ml H2O
10g Rit Dye (Black liquid)
5g LME
10g Agar

Also, only use Rit black. My mentor has tried other colors, but for whatever reason, the black ink is best at slowing contamination.

I use a tiny scoop of activated charcoal in my agar to make them black and give nice contrast. I presume it's not nearly as bad to consume either.

You can send rit dye agar straight to grain if you want. You won’t end up with anything toxic to you in your mushrooms.

There's more to life than cubensis.  :smile: 

Plenty of other species give better trips than cubes and you don't need as much weight per dose/grow either.
