You should be fine to leave your no-pour agar in a cupboard.  I keep mine in a shoebox.

It should be usable for approx 6 months depending on your ge hole. 

If the hole is bigger you will lose a little more moisture from the agar.

If your dish has gas exchange via micropore
tape, sfd, etc then it will dry out
eventually depending the thickness, surface
area size, type of gas exchange filter, etc

I use 1/4 pint widemouth jars with sfd grain
lid setups and they last about 4-5 months
Before they start to dry out. They're fine
to store at room temp before inoculation
for a couple months even if necessary.

With no pours I usually put them in the fridge. They keep for months. I’ve had no problems using plates that’d been PC’d and then refrigerated for about 2 months but I’m sure they’ll last MUCH longer

Obviously this isn’t best practice and when you start seeing the puck visibly dry and shrink you’re gunna want to dispose of them.

Storage in a cupboard is fine for at least a few months or so I’d say.

If they are sterile, which they should be, then they’ll last months in the fridge or at room temp. They’ll last longer in the fridge - they’ll dry out more slowly. But if they’ve been poured and sterilized correctly, you won’t see contams from them sitting around.

Until they dry out. How long it takes them to do that depends on many factors like the RH of your room, whether you left them without wrap vs used cling wrap vs used parafilm, etc, how thick you poured them, etc. The bottom line is that if they didn't dry out, you could leave them for a decade... doesn't matter. Cling wrap prevents drying better than parafilm, but even parafilm works for a long time. Cling wrap has the advantage of being much faster than parafilm in that you can stack 10-12 plates and then wrap them with a roll of cling wrap in one movement. When done like this, the plates will last many, many months.

Quick question:

Once a contam like trich presents itself in a tub, should the entirety of the tub no longer be considered a candidate for tissue sampling? It started pinning yesterday, and this morning trich showed up like a magician put it there. I’ve isolated the tub and I’m going to let it do it’s thing. I’m sure it’s a lost cause, but would you recommend taking a pin and putting it to agar or wait for another opportunity?

I've taken pins from contaminated tubs.
Actually working with 3 different ones
from different contaminated tubs.

Just gotta clean up on agar with transfers

Ok excellent. I’ll take a pin today and put it to agar. Hopefully this one works. Haven’t had success with agar yet. It moves really really slow and then stops. Transfers never do anything. I’m thinking a pin has a lot higher chance of success

Edit: sorry it’s not 10g of each dry ingredient, it’s 20g. Sleepy math.

That’s what I was thinking was the most probable cause, especially with the mycelium I’ve cultured, since it wasn’t from a clean source. Is it usually difficult to clean spore syringes? I have some that are growing, they’re just growing very very slowly.

You using a flowhood or SAB?

Could also just be your technique.
Are you cooling tools down or letting them
cool down all the way before touching them
to tissue or spores?

SAB, minimized airflow in apartment for about 30 min beforehand and a spray of Lysol. All work surfaces and tools sprayed/wiped with 70% iso and allowed to dry. SAB wiped with iso and allowed to dry. I use scalpels that are individually sealed, handled with washed then gloved then iso’d hands. I flame the scalpel then press it into the agar to cool it, which is shown in one of the guides here as an acceptable method of cooling. I cut a small piece and transfer it mycelium down to a new dish, being careful not to open it until that moment, not hover my hand over either plate, and then quickly lid it. Label and wrap with an inch wide piece of saran wrap. Then it goes into the cabinet with my grows on a separate shelf with the same light cycle and temps as the others.

I think that’s my whole process. Do you think the tools are too hot and damaging the mycelium when I transfer them?

Well it’s growing quickly. I’m obviously dealing with contamination issues since I don’t have a clean culture yet, but speed is not an issue with anything else. Even bacterial contam doesn’t really slow anything down. It’s trich and other stuff that’s getting tubs yeeted.

I was dealing with a bunch of plates stalling, I thought it might have been my agar recipe because I couldn't see any visible contam... just healthy looking mycelium that grew for about a week and then stopped...

I ended up trying a different agar recipe and moving on,.. but when I went to look at the stalled plates again a couple weeks later, I could now see some sort of contamination growing.. light whispy growth.. thinking it was some type of mold :shrug: I can post some pictures of the plates at different stages when i get home so you can compare... I'd recommend holding on to your stalled plates and checking back on them in a few weeks to see of the contamination has become more visable

I have a riptide plate that I’m 99% sure is showing some signs of mold. I’m guessing it’s gotta be contam issues, which sucks because I’m not sure how I’m meant to clean them up if they don’t grow at least a transferable section. I’m not going to toss them. I’m sure there’s valuable knowledge to be had. The quest for clean culture continues.

Hi there! I have two grows going, B+Cubenis and Golden Teacher. Everything is going great and I am harvesting right now. I did not label my grow bags after inoculation (my bad) so I don't know which strain is which.  My question is, what are the main appearance differences of Golden Teacher and B+ Cubenis? One fruiting chamber, the mushrooms are longer and skinnier with a pointy top caps. The other container, my mushrooms are shorter, round smooth caps and VERY fat.  I am harvesting right before the veils are opening if I can catch them in time. Both are golden brown in color with white veils, grey gills for those that have opened. Thanks!
