Last summer I let 2 jars go probably a month (maybe a little more) after full consolidation and they fruited. It was new genetics for me though, the flushes on both cakes were unimpressive, but like I said Iโm not sure if it was genetics or because of the longer shelf time

Quick question?
Have just got my ingredients and pots for starting my agar adventure, would long tweezers and a craft knife suffice for tools needed? Thanks in advance โบ ๐๐ผ

As long as you sterilise or at the very least sanitise them before use you should be okay. I used a craft knife for my first 6 months or so.

So give it to me straight. Am I screwed? Should I throw this brick an try again or is there hopes to save?
Everything was great till my first flush. After I harvested for the first time. I soaked the bricks in distilled water for 12 hours each. It's been a week an they look like this.
For the record. Total noob here. This is my first attempt. Appreciate all the help.

I'm not sure if they can be saved, id wait for a second opinion,. but if that picture is in your fruiting chamber you should cut back your foil. It should be barely larger than the cake, that way the perlite is able to do its job properly.

No problem.. I'm not positive but I think that's part of your problem.. it looks to me like it's gotten dried out and matted.. but I'd wait for a 2nd opinion to make sure..
My recommendation would be to start your next project right away... it helps to keep a constant rotation of new projects while learning. That way, if things do go down hill on a project, you're able to just toss that project and not worry too much. Its a lot better than having to toss your only project, which really sucks.

Yeah you can try and save them. Like rotpin said cut the foil down and raise your RH by misting the walls of the tub heavily and probably watering you perlite.
It looks like a good pin set and the nasty looking cake parts seem like bruising and matting because the cakes dried out.

Question: do you always dip your swabs in sterilized H2O before stabbing plates, or do you just do that with older spores?

I only hydrate older spores, swabs are streaked and stabbed.

I seriously doubt you have a monoculture, just nice uniform growth is what you're after, rhizo is popular because it's easier to see there's no intertwined nasties, and usually means it's quite vigorous.

Some examples......
This is a T1 from multispore. There are obviously several sectors here.... several cultures growing on the same plate. To my eyes, this looks clean, but there is some risk there - perhaps the area growing around 9:00 is contaminated by some other fungus. Again, it's probably clean but some people (including myself if I've already tried growing a variety out and had contamination issues) might take more transfers until they don't see sectoring like this:
This is someone else's photo. I post this here to give an example of bacterial contamination. You can see it on the bottom. Note the sharp edges and yellowish color vs the fuzzy edges and whiter color of the mycelium above. Bacterial contamination is often round.

You want to avoid anything of an odd color too... small black, brown, green, etc spots are contamination. I don't have any pics of that handy.

When most people say "monoculture", they simply mean "even, consistent looking growth." A true monoculture is one dikaryotic connection between two spores. You can't take transfer after transfer and be positive that you only have ended up with one true monoculture there. But unless you are trying to grow crosses, it doesn't matter. You don't need a monoculture. You just transfer until you have nice, even, fast growth. Just remember that transferring is a double edged sword - you don't know if the culture you end up with is going to fruit, or fruit well. The time and energy you spend transferring could be wasted if what you end up with is a culture that never fruits or fruits poorly. This is why I usually err on the side slight contamination risk..... I would send plate #1 from my first post to grain, which is a T1. I rarely go beyond T1 unless I see really ugly looking growth like plate #6, which I would not send to grain. I want the widest genetics possible in hopes of stumbling on something good or unique and then cloning/printing it, understanding that I have a bit more risk of contamination (always fungal in my case since bacteria is so easy to spot).

How long is agar (no pour method) ok to leave before inoculation and are they ok stored in cupboard?
