Guys, these dots looks that contamination to me... what do you thing? If that's the case, should I transfer them? I was hoping to not go past T1 for this one.

They might be satellite contams hard to tell. The mycelium near them isn't good anyhow to transfer. You want to take from that 1 o'clock to 4 o'clock region on the other side anyhow.

my filtered NY/LME/AGAR is clear and the myc is liking it...



Looks like I might try to practice LC which means I'll need cool modified jars and have to test it on agar cups...

I'd buy some media bottles for LC. Even though I also use half pint regular mouth jars with whatmans I prefer pouring out of media bottles. Unmodified lids.

SSLC is dope but its much harder to tell if you have any bad stuff going on since the mycelium stays a little more broken up and in suspension. It's also cloudier.

I say all that to say I think soft agar is superior to LC. It colonizes grain just as fast as LC. It will coat the grains instead of settling to the bottom similar to sslc, and you can identify and transfer away from contams with it. On top of this it also doesn't mess with moisture content of your grain.

I'm not saying don't try LC. It's great especially if you can pull it off consistently(I still mess it up all the time) but I am saying maybe try both LC and soft agar and see which you like better.

I know you are doing bags and LC is probably better for going straight to bags, but if it was me I'd go soft agar to grain master. Then grain master to bags.

Or not idk. You'll figure it out I'm just giving my opinion on it even though I've never had the pleasure of running bags.

Hey guys, what do you think about this (apart that they are crappy photos) it's a T1 or T2 of MSS. (First time agar)
Seems like it's 1/3 tormentose and 2/3 rhizo.
Or is it contam? Thanks!

Looks like tomentose and rhizo to me. The long reaching out tendrils could maybe be a sign it's running from something. Or could just be genetics. Transfer some rhizo see what happens is all you can do.

First time with Nats, and after plowing through the official nats thread, I saw the potential difficulties with overlay and fruiting. So after I made some transfers of a MS agar plate, I shelved the plate and hoped for pins. Well here is the plate now...

I have a (likely dumb) question about agar and BFR jars. I know BRF are primarily meant for MS and LC. I have a bunch of T2 and T3 dishes (some that did not make the cut for transfers compared to others) sitting around doing nothing except the ones that are getting ready for T3 and T4 transfers.

With the ones I am no longer transferring with, can those be cut up and put into sterilized BFR's? Or am I just asking for contam for opening the BFR jars (even with my SAB)?

the rules of the SAB still apply to BRF jars. you can open them without contaminating them. there are a couple reasons its goofy to inoc BRF with agar but if youre just playin around yeah go for it

Curious as to the reasons it's goofy, please!

And yeah, this is mainly me just goofing off (and trying to learn more), but also trying to see if I can get some prints from ones I don't have any MS or LC for that seem to be having issues with in my current PF Tek FC.

baked might have more reasoning to go with it but a big part is probably that BRF is more useful for cleaning up bacteria & contams or for germinating spores than it is as an agar substitute if you’re already getting solid growth on an agar plate. but BRF will pin really readily if you leave it long enough because of the nutrition (plates will also pin but can take a long time ime) so you can get good clone candidates that will presumably be sterile & can let caps open up to get a few prints that will have a clean edge over fruits that were fruited out in the open vs. in vitro

I'm not using it as an agar substitute, mainly this is me goofing off, trying to learn more, and that I have a waste not, want not personality.

In vitro is growing fruits in the agar plates, right? Making sure I'm remembering correctly.

well yeah, that’s fine you’re totally free to just goof around & try things, but you asked why it was goofy lol

yeah, in vitro means β€œin glass” or something similar im pretty sure. since it’s all in a sealed sterile container, that makes anything in there also sterile (assuming you’re only opening it in an SAB & there aren’t any contaminates that snuck in)

So, sorry for the shit pic. I'm dead tired and lazy.



There is germination underneath the swab (you can't see it on that bad pic). There is mold on the left. I expect the germ to be another 2-3 days before it's big enough to take a transfer, but I also expect that mold to shoot spores anytime now.

Once the mold goes green, what are my chances of salvaging a clean myc transfer from under the swab? 50% or 0% ? I never had to attempt a transfer from a plate with mold on it.

I have no clue if I just got lucky when I did this, but when I saw mold on a plate I was trying to save, I took a small amount of scotch tape, opened the agar, and sealed the mold. I was able to save the germinating myc for 3 days, and get a transfer to another agar.

But like I said... I may have just gotten stupidly lucky???

Very safe to say my cultures are enjoying the added gram of nutritional yeast I put in my plates. These are T4 mazatapec cultures that were put to grain at T3 then marked for observation. They’ll be going into cold storage in a couple of days.



Edit: the T3 plate for comparison.
