My first FP+ swab didn't start and i wish I'd stabbed it in the dish. One swab of it left and I'll definitely be doing that.

Call me the anomaly here, but out of the 3-4 plates that I swabbed with APE the one that I decided to stab was the only one that had a mold party.  All of the other cultures were/are duking it out on my plates. 

Did a couple of Josex pokes and a couple of transfers (that will be JP'd as well too) but no more smash and leave when it comes to swabs.

Is it normal for a plate to temporarily go more aerial/tomentose temporarily right after opening it to do transfers, and then going back to rhizomorphic?  I'm thinking it could have been from the change in FAE from sitting in front of a hood for a couple minutes 🤔 :shrug:



The areas that look light on the right of the 2nd sample had transfers cut out.

I have definitely observed that. There was also some discussion a little while ago about the burst of fae being a plate pinning trigger sometimes.

Awesome 👌 I appreciate the reassuring response..

I had a feeling it wasn't stress or contamination because the transfers and test jar I made are looking clean, but I just wanted to double check 😅

Also, that's interesting 🤔  I've got a couple plates that should start pinning and time now.. maybe I'll pop the lids open for a minute and give them some FAE

Now that I’ve got a couple of stacks of reliable plates poured, I’m revisiting MEA to see if I can make that work. Doing just 100 ml of solution and a few no pours. If those are successful with trial cultures, then I’ll make a move towards that for plate clarity. Have some T1 and T2 plates that are zipping right along. Nothing worth taking a picture of, but there’s visible growth in under 24 hours. So if all else fails, I have BRF agar that works in both poured and no-pour plates.

If you have another sterile blank swab, I’d recommend putting a drop of sterile water onto where the spores are and taking the blank swab and rubbing the two together before streaking… you won’t have any problems getting spores onto the agar by streaking when doing that

That's a good idea also thanks. I know it's just how I used the first one is why it didn't start. I smeared and twirled it and agar got on it then I smear streaked that off it across the plate. 2 weeks later still nothing. I'll get the next to work though.

Hi everyone, newbie here! These are my first transfers from plates that were inoculated with LC. I did the transfers five days ago now, and just did second transfers from two of these plates last night.

This is my first experience with agar, so I'm wondering how these look to y'all, and if I did second transfers from reasonable spots.

I made a T2 from this one at the very top. Is it normal to see the ropey growth mixed with more tomentose like at the 4-6 o'clock here? Or is that possibly a sign of a hidden contam?


Another first transfer of the same strain (somehow I got agar on the lid when I poured this plate :facepalm:):


The next two are a different strain (PE) and seem to be growing out a bit slower and less dense on LME plates.

I made a T2 transfer from the very top on this one:


And finally... Sorry for the condensate on this one:


Would it be terribly stupid to try a grain transfer from any of these after they grow out a bit more? Am I being too impatient?

I'd really appreciate any feedback anyone has!

The bottom  plate definitely looks the best :super:  I'd grab some of that rhizo growth you were talking about on the first picture around 2-3 o'clock.. but that's just my 2 cents, others may have some different thoughts

top two need a couple transfers
1st - 2-3'oclock and anywhere 7-11'oclock tbh
2nd - 4-5'oclock looks decently even in growth and not fluffy like the upper half

that bottom one id put to grain no problem
3rd one also might be good for grain i think it is growing oddly at 12 because you put your scalpel into the agar there i assume - if so then send that one as well

edit: im bad at reading, the 3rd one you said u took a transfer from but that plate looks ready to me :thumbup:

I usually take a transfer or 2 from a plate when I send it to grain..

If you do send the last one to grain, I'd still recommend starting a transfer plate or 2 from it

So basically I made a transfer from this plate some time ago because it had signs of contam not sure why but I have been keeping all of my plates even the contaminated ones which is probably a bad practice but anyways I got my first ever mushroom from this contaminated plate. I am so excited and hyped like AGGGGHHHHHH First ever shrooomie even though its super small its super exciting for me!

I suppose I am going to clone it, Should I drop the whole pin onto a new agar dish or cut it up and drop onto a few dishes or I I slice it in half and take a small sample from the middle?

For those of you who know about the Tidal Wave culture that I've been having terrible luck with (it was enmeshed with something that would even ride along with plate pins)



I think I finally got it clean :dancer: but I'd like some second opinions, incase there is some indication that it's still enmeshed with something I should be looking for that I'm missing.
