How long do you guys wait for a agar culture that’s been in the fridge for a few months to warm up and be ready for transfers and sending to grain?

0 seconds

Just send it

With that logic it's gonna "warm up" on the new plate or jar anyways lol

I’ll admit I’m a noob, but 0 seconds worked for me multiple times. Both A2A and A2G. I have waited 24 hours to let grain jars warm back up before going to bulk, but I’m skeptical that’s necessary.

I take a transfer from fridge plates and let that grow out before transferring out to LC.  I've been burned by going straight to LC/grains a couple times when I didn't take the extra transfer.

What does an organism have to gain from 24 hours in one suitable environment vs 24 hours in an environment that’s also perfectly suitable?

That’s like transporting a person from a beach while they’re sleeping to another beach across the world. When they wake up, it’s slightly different and they might have to use different tools but they’ll at least walk around.

I send fridge plates without waiting, no point in storing them if I thought they would be dirty coming out.

If you're gonna attempt a transfer do it now
where it's the furthest away from a satellite
growth, and take a very small sample if possible

I prepped 7 grain jars last night and inoculated them this morning with these FP+ plates. I could not see any contams, but plates grew out in basically two different ways as pictured. I don't know what made the difference--genetics, invisible contamination, or what? Anyway, I'm just pushing forward with what I've got and hoping for the best. They seem pretty healthy to me anyway.

Here's all my FP+ plates just prior to transfer.

Please forgive the atrocious pics, camera's busted. But I'm just trying to figure out if I should be taking transfers from these germ plates at this stage?

The second plate germ'd first, but there are flecks of contamination starting to encroach on its margins. The first plate seems to be totally clean, fingers crossed.

Do I let em grow out more, or do I start doing transfers?

If the plate looks clean, I'd let it grow a bit.. but you may want to wait for someone with more experience to reply

Is the one with contams already sporulating?  If so, I wouldn't use any cultures from it since you have a healthy one available.  You can transfer any time after you have clean growth away from the inoculation point.  Letting it go longer makes it easier to find the faster cultures, but you don't want them to get overgrown where you can't tell what you are transferring.

I honestly don't know if the contams are sporulating, as I'm not sure what they are and they wouldn't show up on my camera. It's some kind of film that is slowly growing little colorless specks that show up under bright light fairly evenly throughout. One spot is larger, not sure if it's a different species, but it is maybe turning yellowish in the center.

I'm not too worried about a transfer contaminating as I have lots of agar, rather I'm just trying to move things along as fast as I can, and have the eggs spread out in many baskets xD

On that second plate, the protruding myc is farthest from contams. I'm wondering if I should transfer asap to avoid bringing along unwanted visitors.

I'm pretty sure the contam was spread around by heavy condensation, so there is quite a bit, especially in spots where water pooled.

Mainly I just want to make sure this is viable for transfers, or if I might fuck things up by jumping the gun. Also, there's like zero chance that this is monokaryotic, right?

How do these look? Should I pull a transfer or let them grow out a bit more. Do they look clean? If I did transfer, on the 1st plate I'm not sure of where to take from. For the 2nd I was thinking 3 & 9 o'clock but please correct me if I'm thinking wrong.

"Hella tome" ........yep

Thanks for the encouragement and good luck with your FP+. Mine germinated fast and contam-free. Praise Faht, the man sends clean swabs!

Can anyone tell me if the way this myc grew out is acceptable or bad, and why? Thanks

Depends who you ask from what I’ve seen on here.  That area appears to be colonized but with very thin myc.  Nichrome says it’s just sectoring and under a microscope it appears to be myc there.

Id imagine it’s not great to have a bunch of that myc in your grain jars tho.  Since it colonizes so thinly, maybe it’s not great to have that myc on a lot of grain kernels as it doesn’t provide very much protection against an aggressive mold spores/mycelium.

If it was bacteria, it’d grow like bacteria and look like a booger but that’d be a cool project for someone to figure out definitively

But I’m just speculating.  Maybe someone that has experience with sending plates like that can chime in

I've never sent one myself because it "doesn't look right," but love to see what it really does.

Ah, thank you very much Faht. That's reassuring. I searched Nichrome's posts and found it right away   So this might just be sectoring due to genetics. I hope so. We'll see....

Thanks for the feedback Sandman & Mushboy, too. I will track this plate to grain and to harvest and we will see how my grain jars and tubs eventually turn out compared to jars and tubs that started out from plates with more uniform growth. So, now I have a plan anyway
