Given the low price of 100 sterile single use scalpel blades, I change them every time I use them. Sometimes if I’m doing different kinds of blade work, like taking tissue samples from a fruit and then doing agar transfers, I’ll swap the blades out between tasks.



I do the same. As long as I’m transferring from the same plate or fruit I leave it on, but when switching plates or fruits I typically change them. It feels a bit more wasteful than I’d like, but at the same time I’m also not doing large scale growing so it’s not like I go through a lot of blades that way either. It’s pretty convenient.

Pretty much when it’s too dull to cut the agar accurately. Few months for me.



I do it once they start to looke like they've been flame sterilized one too many times... maybe once every 2 weeks.

I usually keep 3 scalpels in rotation while I'm working, so 2.weeks is kinda quick.. but I flame the hell outta my blades 🔥 I could probably get away with keeping then in the flame for half the amount of time that I do.

How long does it usually take you guys to flame a blade? Do you just let the blade flash red for a second, or do you let it stay red hot for a few seconds before removing it?

That’s when I change it, when it’s just too warped and blunt. Usually about every couple months. No real need to change it much more often than that. That’s me personally though.

These yetis looking ready to harvest? There are a few aborts mixed in with them. Some veils are tearing. Some do feel kind of marshmallow like. Some are having blueing appear of the caps around the edges. They all appear to be apart of a few clusters which means if one goes they all going. I was thinking maybe waiting till tomorrow?

If it turns red it's sterile. If it will boil agar it's sterile. Thicker steel you don't even need to get red. steel shouldn't be porous enough to need to get all the way hot in the middle. With flames you are also dealing with a lot more destructive power than just heat. Fire kills things quick.

Some folks out there flame their tools so hard you'd think they were trying to re-forge the blade. Seen people say their scalpel handle gets hot from flaming their blade

I'm not quite that bad, but I've been keeping them cherry red for about 3 seconds. I'll probably start cutting back.

Most lab practices call for just a four or five slow swipes through a LP bunson flame.

Ethanol burns optimally in regular air at 3779 F. That's what I use in my alc lamp. If you are using a gas torch it is considerably hotter.

Rotn, that's probably a better description of what I'm doing also.  I try to keep red-hot afew seconds but am trying to be as quick as possible too.

Quick question, for anyone using mini rounds/pastys, how long have you kept plates in rotation?

Right.. I may have to start some counseling or meditation 🧘 I've got a bit of a germ phobia, and this hobby hasn't exactly done much to help that.. I think flaming my blade for a shorter time period may result in some anxiety issues..

Test plates, combined with critter brain reset, should result in a no fear /no anxiety scenario. Takes a few times to develop a comfort zone.

Yeah, I agree.. better safe than sorry.. I like knowing for a fact my blade is clean. since you're using a SAB, just use a cooling plate..

Working in front of a hood makes it a lot easier.. I just flame 3 blades and set them on a jar to cool while I unwrap the plates I'm transferring from/to and then start with the first blade I flamed, it gives it more than enough time to cool.

same, but it still happens.  I keep an extra plate with each group that I use to cool the blade before transfers, it hasn't seemed to add to higher contam rates, but there's a decent amount of contam here

If you do a search for "monitor stands" on Amazon, there are a bunch of options.

I'm pretty sure these are the ones I went with.. I got 4 of them (they come in sets of 2) and I took 2 of the legs off of the one on the end so it would fit on my table and give me a workspace across the whole length of my hood
