Clear dish is brf agar after 4 days and I think its doing waaay better than pdya agar. All t1 except last dish is germ plate I xfered from.
I Def need to add some charcoal next time because on some other dishes it's really hard to see the mycellium

My gt germ plate from mss from 3 weeks ago grew a distinct fan area that I thought looked good so I did some new t1s 4 days ago. I took from the full white fan area and also xfers from both sides of the fan in the more transparent areas.

Now it seems the weaker transparent areas are growing faster and more distinct than the fuller white fan area. Not sure what this means. Maybe the white area wasn't as clean? Or is it just more tomatoes?


Pics are bad I know, hard to get a clear shot of the one with condensation.

Also, the TAT plate that is producing strange blobs and other odd variants with plates made from transferring some of those growths:



In addition to blobs there are some squat dwarf-pins reminiscent of APE that aren't very visible there.

If you got your subtrop from the same giveaway I got mine from, looks like yours is a lot like mine.

I received subtropicalis prints from two users, A and M. I believe that plate was made from M's but both are currently in use. A's print was allegedly very old so I decided to give both a shot. Neither were from giveaways. Both sources produce similar, textbook subtropicalis mycelium that is also doing very well on oats currently. If either of you guys read this, thanks again for giving me the opportunity to work with this species!

That looks great! I never take quite such clean shots of petri dishes.



This is a T1 originating from some of your plates' spore brothers. Ignoring the battlefield that is this plates surface (black blobs are just from molten agar produced during a messy hot straw transfer), there are two distinct mycelial sectors here it seems so, as you can see, I'm trying both out. It could just be some environmental factor causing the different appearance I suppose. Further work will tell.

Okay, so I grabbed a small cluster of two pins from my little Atl 7 grow, dropped them in sterile distilled water, shook them, dumped them out in front of my flowhood onto a sterile pp5 plate lid, squirted hydrogen peroxide into said lid, let sit for a few minutes.

Dug pins out with a sterile scalpel and then cut several pieces into a four separate plates.  Trying to smash the pieces down into the agar as much as possible.  Probably 8 - 10 pin pieces total.

Plan is to grab the edges of any clean growth I see and transfer to a clean plate.  And maybe do one more transfer like this just to be sure.

Are the odds of this working okay?  Or am I going to end up with hidden bacteria in my clones?

This is my first time trying to clone shit this small.

Can someone please give me some advice? I've bought 1 print and 1 syringe, from supposedly reputable sources, and had 2 prints sent to me from shroomery members. Every single plate I've tried has developed these thick halo looking rings around the myc, and I've made like 20 plates and the best looking ones still have this weird growth and a thick halo ring.

My PE culture I have cleaned up and I regularly clone tissue, go Grain2Agar, and have successful monotubs that pin nicely. I also pour way more plates than I need normally, and none of my plates contam except the ones that I streak.

IDK if these halo rings are normal on the 1st xfer, and not a sign of contam? I've made over 20 plates of each strain, and not had ANYthing that looks xferable. The BEST plates have these thick halo rings, and seem to have a ridealong parasite.

I'm so tired of getting prints and syringes that don't give me anything remotely healthy looking. Can anyone PM me and point me in the right direction? I know how to do agar work, my PE works great, and I've cleaned cultures up before. I just can't seem to get a clean, regular cube print to save my life. I really, desperately need access to a print/syringe/swab that will give me some myc without parasites. I can deal with bacteria, these ridealong parasites I CAN NOT seem to get rid of.

What could I possibly be doing wrong? I know these plates all look terrible, but these are the best I've gotten and I've gone through at least 100 agar plates. I'm desperate for a regular cube culture and I feel like I've wasted so much time already.

That halo on that second and fourth plate I always thought was just new growth.  And that the white stuff that's seen by the naked eye more easily just takes a bit longer.

That's just my theory though.  Could be wrong.

I agree

In regards to manlords plates, some plates are harder to clean up then others. How many transfers have you done? Id keep transfering IMHO. Ive had plates take 1 transfer to clean and plates that took 7. Wana see a real dirty plate?
