Once inoculated I’d wrap the plates.

The sides are wrapped with grafting parafilm. Did I wrap them wrong?

Sounds like my sab pour rate. Every couple stacks one or two would mold out. Chucked it up to collateral damage. It's a sab it happens:shrug: I have to wrap my plates after pouring now. They mold if I dont:rolleyes:

I like the white marker what kind??

Any advice on cloning very small fruits?

I just tried to clone a couple of Tamp fruits and it didn't go great.

Was considering using the tip of a hypodermic next time to scrape the insides instead of a scalpel.

Thoughts?

Go for it. I just mashed some piece of stem into the agar and waited for something clean.

It's cool if it's just a thing that happens, I'm mostly just trying to figure out what to do next. Like should I wait longer on the swab or maybe transfer that tendril coming from it? Also, the Amazonian plate, is that... all bad? I just don't know enough yet to be able to tell.

The pen is the silver from the Bic Intensity Metallic Marker Series, nabbed straight out of the bowels of D3's Perfect transfers and agar TEK, which is what I follow for making plates. (Thanks D3monic!)

Also, excuse me while I fangirl a little bit. Love your posts, too, mushboy. :mushroom2:

I posted these in different thread didnt get much of a response .
I’m pretty new ... How do they look ?

I know this one is no doubt looking good
Ape clone 3rd transfer

But what about these
Ape clone 4th transfer

I included close ups and flipped over a lot of my samples have an orange tint .. but it could just be the reflection of the colored agar..

And how about this one .. just weak looking myc in my opinion .
Ape clone 4th transfer

And same with these with weak looking myc

These are Amazon 4th transfer

I was having issues for a min with plates in my flow. I had to go back to letting my flow run all night and wrap the next morning in the flow. If I poured and turned it off and wrapped the next morning or evening I would have a few contaminate.

No, just wrapped so well I couldn’t see it. The couple with edge contams make sure you don’t have any splits in the parafilm. Happens sometimes if you stretch too far it splits at the seam between two plates.

Bacteria from a swab not a lot you can do, try to keep the wood off the agar if you can or pull little bits off with sterile tweezers and dunk just the cotton

Oh, okay, so you think that part has bacteria. I thought it was the blob on the side of the dish that was contaminated. Thanks for the advice on the parafilm, maybe I'll start double wrapping.

The ooze leading out from your swab in the swab tip close up def looks like bacteria or very similar.

Para: The streaks in your first photo are likely from beads of water picking up bacteria from a smaller contam spot, then spreading it around the plate when you picked it up and let the water run. But I see a spot that could be mycelial growth in between the streaks of bacteria. If it is, you'll want to transfer it ASAP to rescue it from the bacteria.

Your other plates could still germinate, but I expect to see visible growth in four days if the spores are weeks to months old and in good condition. If they were in storage, it may take longer.

The white circle on your 2nd image seems like it grew too fast to be mycelia. It's been a week since streaking, no? As for the cotton swab, you can wait. If it has bacteria, the mycelia may outrun it. If it has mold, the swab won't yield useful samples. (If you get nothing else from these plates in a week, there may be ways to clean up mold but I don't know how.)

Thank you very much, pear!

I think I'm just weirded out by the first plate because they haven't been moved at all or tilted until I moved them out for a picture. The growth doesn't match my streaks either and is actually running against it. That's surely bacteria like you said. I'm sure you're right that it's probably from drips or something. I'll definitely try to transfer from the spot between the streaks if it holds out until the morning.

For the plate with the swab, the circle on the side is surely contamination, or at least unintended. Should I try to remove it since it's on the side?

I apologize for potentially asking questions about things you guys take for granted but I'm never sure the best way to approach asking things on this site and sometimes get a bit of posting anxiety.

I really appreciate the help very much, though. Thank you.

No worries, Para. But don't transfer contams away from mycelia. Transfer mycelia away from contams. Perhaps because mold can easily spread by mycelia fragments and spores, so it's hard to remove cleanly.

Para: for some types of contam, you can pour more warm agar on top, and only the mycelia will grow through. But I'm not sure if that only works with antibiotic agar... and maybe mold can also grow through it. I'm not advanced on cleaning up cultures.

Kuda: TL;DR: they look fine, some could use another transfer.
So many pictures, so hard to give them the attention they deserve. Your first one seems to have more than one strain fighting it out since it's not symmetrical. That's okay, but it means different spots may have different phenotypes. The "edgiest" parts may be fast colonizers, but the truth is you really only know that they are fastest on agar. Still, there's correlation. Your middle photos look similar, which is good. It's interesting that mine don't look like that, but that just goes to show that different varieties can be different, even on agar. But they look fine. Your "weak" plates look--but it's hard to see--like linear growth. That's not bad as far as I know (but nobody has made a numeric study of this). I've grown from linear plates but not from the "fuzzy linear" plates like your middle ones.

Keeps them shut. So far it seems to work as well as parafilm with less effort and money.

I keep plates in zip lock bags, about 15 in each gallon bag, and keep the bags in a box in an unused room.

You may experience satellite contams. The wrap also keeps ickies from getting up under that lid.

Edit noticed:typing:
