agar to agar transfer

ey!

since i got my first petri ready for a transfer to new ones,
i searched the shroomery and read whatever i could find.

from all the stuff i read i came across two common practices.

one is to simply cut a piece of the colonised agar and drop onto the middle
of a plate, then seal.

the other is to cut three different little pieces and place in equal
distance to eatchother on the plate.

now, i see that that the 3 'inoculation points' are going to give the
new plate a way fater colonisation, but i wonder if the one piece in the middle
method got any disadvantages?

im somewhat limited on my agar, so i dont have too much to toy around:

i am going to use peroxided agar plates.

the above shown plate was from a CLONE, i extracted mushroom tissue and placed it on the plate.

Number one isolate isolate isolate! Once you have isolated a pure substrain then you could use the three point method but for now you can take a small piece no closer than 1/2" from the edge this will help keep contam spores to a minimum but also remember that the farther away from the initially transfered isolate (placed in the center of the dish) the purer the myc will be to a point. If you have only one plate and it was from a transfer from a multispore swab then I'd take three sectors and isolate to three plates, compare there growth characteristics and decide from there which you'd like to use. I always recommend one section and placing either in the center of the dish or off to the side to allow for easier isolation of pure stock. GL

When trying to get an isolate, I will transfer more than one piece onto a single dish. For instance, if the source dish has some myc. sectors, I may take a piece from each sector.

After the strongest growth can be isolated, that's when I transfer only one piece in the middle to have a full plate of that isolate - used for inoc'ing sub. or putting up for storage.

thank you for the input.

the plate above is from a cloned mushroom,
in the center u can see the tissue extracted from the stem of the mushroom.

i am going to follow your advise and cut very small pieces of agar and
place one little piece into every petri, ill do about 10 petris this way.

also, if i still got remaining agar, i will transfer some to '3 pieces petris'
and let it grow out.

if you have got any further suggestions for me, please post.

Since your working with a pure isolate already then if the plate was keep sterile then you can use all the agar even right up to the edge. Now I assume your building stock cultures since you said your going to do ten plates, thats alot for a single strain and really not wise IMO. I would make say three save the rest of the plates/agar for future isolations otherwise you will have to definitely use all of these in the next 12mo. for best results. My suggestion would be three plates and the rest should go to grain, then the fun begins, G2G!!! Keep 1-2 masters in the fridge for backups, you'll be over your head in substrate in no time at all bro! GL
