One question regarding SAB, the correct is to mist the SAB first then put everything inside right? Just took a look and another unused poured plate had mold, so im 70 - 80% sterile...

OBS: SAB used to be an fruiting chamber, i closed those with plastic tape =]

I quit spraying the inside. I simply wipe out with iso sloppily before setting the SAB down  (I move my sab around from use to storage.) Iso hands.
Place work inside.
Iso hands then get to work.

Question! Do you keep your lid on the tote and place your work on that? (Hard to tell if the lid is on or not, sorry)

If so that could cause a problem maybe? I noticed setting my work on the lid caused the lid to compress and or snap back to it's original memory causing a disturbance of air within the -not so- still air box now. Throwing contams around with agar is no good

Hopefully this helps someone else out if anything!

Lol, snappy lid got me before. I still work on it though, just don't put weight on the center.

I think being above the actual lid is a benefit iirc. I put plates on empty plates when working. Others have racks. Something bout air moving more near surface?

Here are some updates on the transfers I made last week. I took much small pieces from these, about 14 transfers in total. I like how the malabar and ecuador are looking. The GT is more uniform because it's a transfer in from a clone.

the PE (they're bubbles) and B+..well let's just say I hope they find their legs. And after streaking 8 plates, I have some bisporus showing up finally. Still nothing on the cambo.

Tampa, ATL 7, Bisporus

PE and B+

Malabar - some of the thicker ropier growth is actually lifting off the plate!

GT transfers

The best of the Ecuadorians from the bunch

So far all of my transfers are recovering well. I am hoping for some good uniformity, I will make another transfer and use the rest of the plate to make 1 - 2 quarts of spawn to start making shoeboxes...hoping for something good to clone. This is fun stuff!

Thanks for the info. I keep the lid and try to work on the rack. My container has openings in the side that are just closed with the lid, where the lid holds. If i don’t use the lid small open retangulars will be open

Finally perfected my agar tek; slowly wisking agar and LME into already boiling water is what got rid of the flakes

I'd like to get this same clarity from my glass petris but they seem to condensate a bit worse. Usually only the 3-4 bottoms one are clear.

Question of the day:

What's your favorite way to swab plates?

Do you swipe the plate with the swab tip or do you tear off pieces of the swab tip and lay it directly to the agar?

Just curious to see what the general consensus is.... :awedance:

I do a combination of the three main methods. I streak a few plates rolling the swab as I go, maybe tear a little off of it for another plate and a final dunk plate. Especially if it’s been a stubborn to germinate strain.

Swipe. I figure it's closer to an innoc loop and spreads things around. I plunk a piece if i have trouble getting germination with the swipe.

I like to swipe the swab on an edge of the petri and then use a loop to do a serial dilution for the spores already on the plate :havesomescience:

I couldn't get clean growth just swiping with my APE swabs recently so I just cut off the tip of the swab and jammed it into the middle of the plate and got some good growth.

I always tear off a piece of swab and stick it on the agar. I don't prep softer agar for germ plates so just swabbing is inconsistent for me.

How tight do you guys keep the lids on your media bottles? I just got mine out from last night and 3 out of the 7 were almost a full rotation from being completely tight. I always double check that all the lids are loosened about half an inch before I pc so idk how they got so loose let alone why only half of em did. Its been colder recently and I pc in the garage so maybe that could have something to do with it?

Shits frustrating man

Hey guys.. did spore syringe to agar with AA+ a guy sent me.  Only one plate had any growth.  Take a look - does this look OK to you or is this not myc?

Also, that divit in the center is the hole I made..

That’s a tough one.

It looks like myc just under the surface. I’ve had growth like that do fine after a transfer before, that’s gonna be the only way to find out probably.

Is that plate made with soft agar?

Thanks for the reply. I'm not sure what soft agar is - I just followed the regular agar tek (Bhod's) using agar powder & malt extract.  It's worked well for cloning and other spore growing.

Standard half strength is  2%  Agar, 1.5%  LME -C10
SAGARI .7% Agar, 1.3% LME - CapsMcgee

Sagari is super soft, for A2G, agar-to-grain...

Yeah just any recipe with less than normal amounts of agar which helps with spores.
