I put mine on a stack of 5-6 plates with an iso soaked paper towel over them.

I’ve been thinking about hanging fishing line with something sticky at the end inside the SAB so I could just hang the lid right side down the whole time lol.

I haven’t had many problems using the iso towel but it does feel weird setting the lid down and putting it back on.

Can do a one hand motion too, just lift the plate like 1.5-2” holding with your non dominant hand, then let the bottom half of the plate lower down so it’s open like <  takes a few seconds of practice but works ok.

I've been in the habit of just opening the lid and angling it so that the plate is still covered for the most part. I just try to be quick

Ok! cool I'll try the one handed clam plate trick next time I'm transferring!

It seems to be much easier to cut wedges when holding the plate in a steady position.

Previously when I did transfers I always tried to touch the plates as little as possible. When cutting I was holding the plate down with only the force of the scalpel. But it doesn't always work out and the plate kinda slides around on my steam rack so I sometimes kept it in place with one finger touching the side of the plate.

My hands happen to be just big enough to hold the plate and lid in one hand while I make cuts with the other. Kinda like this.



Works with regular dishes too. 60mm plates are easier than 100mm

I place my mother plate lid face down on an alcohol wipe.
Sometimes its a clorox wipe that got dipped in alcohol.
I firmly grasp the lid of the new plate with a gloved hand and quickly place my wedge.
Can't wait till I have a flowhood.

I feel bad for SAB guys, you’d throw your scalpel if you seen the shit we get away with in front of a hood.

I was looking at these as a punch idea but I think even the largest sized one might be too small if I’m reading right. Somewhere around an 1/8”  I prefer 1/4” pucks personally

Is it a bad idea to throw this whole plate to grain if I have three transfers on it ? Not sure if I can get it to look much better and was just gonna send it on the APEU . I’m gonna make some more transfers on the PESA

I tend to take slightly sketchy plates to grain just to be sure.

Looks good to me. What are the benefits of dropping the nutrient level? I ask out of ignorance. They seem to be doing fine with your current recipe.

Sorry, it took a while but here is a summary of the agar sandwich experiment so far. I got some strange looking results, and the investigation continues.
So I started out with these plates, all of them were looking a bit fishy, that's why I wanted to use them for the sandwich method


Then I transferred a tiny wedge from all of them to a new plate, put it in a small hole I made in the agar and covered with a clean wedge


After a few days the myc was poking through, mostly on top of the wedge. It took longer until it grew out from the sides, no idea why. I streaked the top of the wedge with an inocculation loop to transfer the emerging agar to new plates.


The resulting growth was quite strange. The myc turned really thin and whispy, quite similar to the growth one obtains from spores. Very disorganized and frail lookng. Some started sectoring in extremely tomentose or rhyzo growth. If I saw them just like this, for sure I would have thought they were contaminated.


These were the transfers to new plates


Not quite what I expected:shrug:
These were all MS from spores, mostly transfer 2-4. Maybe the agar wedge helped to reduce the number of strains and now it shows stronger sectoring? Maybe myc that is able to quickly grow through agar just looks like this?

Are you planning to transfer again from the especially rhizo sections in the last pics? I haven't worked with agar enough to understand what the benefit of a "sandwich" is supposed to be.

So ya, agar got too cool, leftover PP plate from way back.
Poured six days ago. So far, 6 auto-contams out of 25 plates. 24%

And the purdiest contam ya ever saw!  :crazy2:
