That’s fine. It’s hard to cut plate pins ime, usually end up fumbling around a lot more and potentially contaminating the pin when I try to cut it.

nmd_myco those look real pretty, if you're wondering about a2g i'd probs still do at least one more transfer for what it's worth, and yeah i'd probably give each of them their own plate. i'm a hoarder with plates though so that's just me haha

Thanks. I missed this when it was posted. That's a germ plate, obviously. I have no intention of going to grain till i have proven clean results with agar and LC. In cannabis, propagation has always been my superpower.  Never got real hands on with tissue culture because of workload and admin responsibilities by the time we started doing it.... so I am really excited to start working with cultures.

The fact that strain selection and breeding happens mostly in the flow hood, makes me very excited in a quasi-sexual way. Obviously, growing strains out and doing bioassays to pick your winners is a big part of it, but I have a feeling that others will be better at that part than I. not that I won't learn on it hard.

I am a 75th percentile commercial ganja grower, 25th percentile farm manager, 15th percentile HR Manager, etc... Lots of hats. but I am a 99th percentile propagation guy.  If you need to popup a facility and make 10,000 perfect clones that will outgrow other commercially available clones by a factor of 3 in 40 days (in 3200 sf, with 16x128site aerocloners), I am your fucking guy.  Part of me wishes I had spent my cannabis career focused on that alone instead of on vertical models.  There will always be people who need that sterile back-end handled for them.

Thanks. Very cool. I’m excited to see how this performs when it finally goes to grain.

Yea. I had a stretch of contams recently so I’ve been trying to work quickly with purpose lately rather than fuck anything up.

Before

mistakes made: did not flame the cube-tube (tm) between each cut

After:

Pictured: cube-tube agar sampler (tm) $39.95. hit me in the dms. (jk) details posted in the tools of the trade and d3 agar threads.

I did not take all the transfers I cut.  A couple of them were too close to the center of a colony... Very interested on feedback as to my choices of spots to transfer and timing. I figured that by tomorrow, the myc on the germ plate would all grow together and i'd be transfering too many different cultures at that point. Thoughts?

Not unhappy with my first attempt at the d3 tek.  Love the tool that i Walmart-Guyvered this morning. I skewered 4/5 very nicely and effortlessly flipped the disc effortlessly 3/5 times. I will be finding more of a spatula type scalpel to try but when done right, the #11 works fine.

I took 5 off that one plate. I have 3 or 4 more good plates of filthy's PESA... do I want 15-20 T1's?  Do I want more t1's and then be more selective with subsequent plates as I try to isolate?  How many different cultures should go to grain if I am looking for the best culture? Shoeboxes at that point vs bigger tubs? Then select special fruits to clone?

Big advance in parafilm management today.  Pinch and push. Pinch and push. Works amazing with gloves... obviously the way it was designed to be seperated from backing!! Those parts along the factory edges that isn't stuck to the backing...repeats every inch. Just pinch hard and push and voila. Duh.

Anyone mind telling me if any of these are good, and if they aren't, what kind of contam they have? I'm new to agar, and I have troubles identifying contam on agar... :frown:

These are from different batches. I think because my technique lacked in the first ones, the syringes may be contammed.. I've tried petri dishes, condiment containers, and glad mini-tek to no avail. My glad minis seemed to dry out and never grow anything, the rest got black mold/cobweb/maybe whatever else is on the ones in the photos.

Here's a link to the image.. not sure if my post got deleted or what, if I'm doing something wrong please let me know. Maybe it was too big to embed, so here's a direct link

I'm not clicking on an external link.

Go back to square one and find the holes in your technique

Hey y'all, I just finished my first transfers ever about a week ago with GT mss to grain soak agar! A few of my dishes contam'd and I accidentally transfered an agar wedge into two dishes I previously inoculated which did not germinate for a few weeks. Pro bally should of written on then with a sharpie. I have a few questions

1. Which dishes look like the best candidates for transfer #two and, do any of them look clean yet and could go to grain?

2. Where would you select the next transfer from?

3. Is the contaminated dish worth saving as it looks like better uniform growth and less tomentose.

I only have a few dishes left that are poured about 6, and would like to try and inoculate some quart grain jars with the best genetics I can so that brings me to

4. Should I pick the best section from one plate and do a few more transfers with only that one selection before inoculating or try a few spots and do les transfers.

MSS to Agar. Some plates I’ll just blend to a slurry and inoculate some grain.

Some will get sectors transferred

Gonna slurry your germ plates tho 🤔? Seems like you may as well shoot your spores to grain at that point.

This way I know for sure it’s clean and the germinated spores on agar means I’m inoculating with growing mycelium.

It isn’t ideal but I have some plates that aren’t good candidates for sectoring out some transfers.

Oh well!

My germ plates are a mess. Was thinking about doing the same thing when my grain gets here.  The timing should be right.

But like...that’s not what the TEKs say to do... LOL

Y’all on this site are some of the most uppity people...

Let’s just step back and appreciate some progress shots and some future plans. Also please note that there are 3 of 75 plates pictured...

No agar to grain right now.

I do have a few zones that will be getting transferred...

Why are you gonna pop off after showing 3 of 75 plates? I'm assuming those are your best ones and thats why you're gonna use them, so we're aiming there's 72 terrible plates off screen?

Dude was trying to save you a headache but alright. 👍

That first plate looks a lot like some mold that I grew. Thats definitely not myc. Its way too wispy and sporadic. Its hard to tell from the pics but the other two look bacterial. Can you get some closer pictures?

I have an update to the Cover Slide Tek that I posted about here:

https://www.shroomery.org/forums/showflat.php/Number/26981589#26981589

Please read the original papers where I got the tek from in the links that my post above will link, too.

The plate now looks like this:

So the mycelium found it's way out of the trap and looks very nice now (it is pan mycelium). Is it free of bacteria, I do not know yet for sure, near future transfers will show I hope. It looks healthy.

This is the plate the sample came from, very dirty:

Sorry for the bad photos. I have only a 15+ year old little camera and no smartphonethingy either.

why wouldn't i just drop a slide cover on every plate from MSS?

drop, cover, wait? seems like a great way to get a good start from spores... maybe wait till you see germination and then drop the glass as a matter of course.

Wow I’m surprised you were able to get that wedge into the new plate without showering spores all over.

Most of the time when I take wedges from plates with tons of mold I get a bunch of satellite colonies on the new plate.
