What's sketchy about it? What things tell you that? Because it looks normal to me. I thought that tomentose mycelium was fine...?

Typically it is.

The way the rhizos seem seperate make it seem like the myc is trying to outrun something

The fluffy rhizo tips seem a little sketchy to me aslo...not sure why...just seems off.

Hiya...I'm still a noob but I've done a number of runs with PastyPlates (awesome) and have just shifted to plates.

I usually use a PDA/PHA mix using Idahoan Potato Flakes and sediment is kinda normal for me.

Don't have many PastyPlate pics to illustrate but here's a backlit one of a transfer I did to a plate of PHA mix recently. You can see the myc, but the sediment is floating there in the background.

Thanks for the input! I'm not stressing about the sediment any more. To get clear PDA do you need to let the mix settle then pour it off to separate? And is there any practical reason to do this other than aesthetics and the improved visibility that comes with it?

I've enjoyed the no-pour tek, but have ordered some bits and bobs to try pouring. Going to try a bunch of teks and see which suits me the most.

Day 2 of my plates and still clean. I'm not sure where I got it from but in my head if they are clean after 4-5 days then they are ok?

Also I've just picked a metal straw for transfers, seems like a fun little tek.

Yeah natalensis tends to have big myc, my ms plates were up to the lid in some spots. Real weird.

On 9/1/20 I made 1000mL of agar using 10g agar agar powder and 8g LME.


I poured 20 plates with the pink bottle and put the orange bottle in the fridge.


On 9/2/20 I swabbed 10 of the pink plates and made 3 transfers from some blue plates that had just germinated.


It's been 10 days and the spores haven't germinated on the pink plates and the transfers from the blue plates are behaving strange. 1 is only continuing to expand through the original blue section that it originated from and the other two are going inside of the agar and expanding outward at a very slow rate.

3 days ago I made a few more transfers from the blue plates to the new orange plates and they're all doing great and moving along normally.



Obviously, I did something wrong to the pink bottle and I cant figure it out. Maybe I forgot to add the LME to that bottle? One of the pink plates that I didnt use is beginning to grow mold though so I dont know... any ideas?

Hey Doc! I wonder if it has something to do with the color? Shucks I forgot which thread but I recall reading where some red dyes were made using bugs...anti-fungal bugs.

If that were the case, that might explain the inhibited growth on that color, and normal on others.

I usually stick to my favorite colors of blue or green or mix them a bit if I'm feeling spicy...lol

Hey thanks!

I'm still a noob, but I went to Pasty's with my first attempts at cloning.

You can go straight to dishes, and I see a lot of experienced folks recommend it, but Pasty Plates were a good agar starting point for me.

I've done probably 40-60 plates in the past using either the round frosty glad cups, or the squarish and more clear ZipLoc ones. They worked great and didn't seem as "formal" as plates.

With that said, if you've done Pasty's before and are comfortable with that, it's not a hard leap at all to go to plates. In fact, once you've done it, and see how much more you can see and observe, you'll wonder why you didn't do it sooner.

I'd strongly encourage you to try it at least once. You can get 30 90x15mm plates off amazon for $14 and a media bottle for $12.

Big thumbs up! Many knowledgeable and informative posts in the forums.

I've been logging my first try at isolation and plates in another thread and plates have really been nice for being able to really observe in detail what's going on down there.

I still have my Pasty containers and I'm thinking I'll use those to make wedges to go to grain with, since they are reusable and if I'm propagating from a known plate, I won't be as concerned with observing the fine details as much as getting a puck to drop onto grain quickly and easily...that will be my first chance to log some Tiger Drop action :smile:

Hey everyone. It's my first time doing agar and I was hoping for some feedback on how my cultures look. The first four pics are amazonian and the last mazatapec. I just put a couple drops on the plates with a flame sterile syringe, I've since made an inoculation loop for next time. Any feedback would be greatly appreciated!

Check how thick this myc is on these T3 Golden Mammoths.



I'm trying to get it to run a bit, but every transfer from each plate is going tomentose. Got T2's on grain, grew out nice once it hit grain. Same thing happening with my B+. APE is being a dick... but got some decent plates that took off on grains.

You don't need yeast. I use pretty much the same recipe. I do 8g LME instead of 7g. I haven't had any plates make it to grain yet. Theres a few that are starting to take off pretty decent but the rest I keep having to transfer away from mold or bacteria.

I transferred this one as soon as mold started near the edge and kept it around to see what it would eventually turn into. Its getting pretty gnarly.


I thought for sure that this one was gonna be my winner but sure enough, there's mold germinating near the edge again. I cut two small pieces out of the rhizo edge and put them on their own plate. Maybe one of them will stay clean.



Here's a plate that I intentionally contaminated to see what it would do. Lots of pretty colors.


It's always exciting to come back to a plate after 15 hours and see how its changed.
