Okay. I suppose I'm just being impatient. Its only been 48 hours since I swabbed plates and Its bothering me that there's not even bacteria showing lol. I'll forget about em for a few days.

has any one used or considered reusing old cracker dry Petri dishes?



I have a handful of plates lying around from over the past however many months, years etc that look like this.

Still micropore taped, and also previously clean culture.

open under the flow hood remove the dried up agar, and pour again

thoughts?

Germination can take up to 2 weeks. Imo a lot of recipes aren't great for germination, i like to dail back on the agar a tad and up on the nutes for germ plates

Thats exactly what i did to make this plate, a tranfer later it looks great! It wasnt that old, but it should still work.

Thhe donor plate was poured so thin i didn't even get any agar, i just scraped up some myc and transferred it

I think i know the answer to this but i assume you would want to do a hot pour while the culture is the size of a quarter or half dollar, then wait til the second layer of agar gets more fully colonized before transferring? Haven't done a hot pour yet, but hella considering it, i either have a couple cultures with bacteria intertwined or my grain prep is shit [most likely shit grain prep tbh].

Oh damn I thought I read somewhere that less nutes will make the plate colonize faster.

I simply mean reusing the plate itself, the agar drops out like a cracker lol

I poured some reused plates earlier we shall see how it goes.

plates are cheap but ive kept all of them from over the years. figured It would be worth a try instead of just tossing them.

never seen this discussed before

Here are a few that are going to grain soon.. I've been alternating between transfers with dog food agar, malt extract agar, potato flake agar, & brown rice flour agar.

Yeti
Melmac F2
White tailfeathers F2
Ape (kingkong)
Tailfeathers clone4 (one plate has mold ,one has bacteria but usable)

Not included in the pictures....
Burma swab plate & T1
Golden teacher "clusterfuck" swab plate
    The GT"cf" is one of the tailfeathers parents.

So my first attempt at agar work was a huge failure.  I originally bought the wrong kind of agar.  The kind that is for bacterial growth.  I tried adding potato flakes to it and was pouring it in glass petri dishes. Im pretty sure I had a dirty syring to boot. None the less, it was complete failure over and over again.

Ive since made some pasty plates, and purchased some MEA premixed agar. I also got a new MS syringe from a vendor. 

I didnt use a loop, kinda forgot about those.  I just shot a drop out of the syringe on the agar in an SAB.  The needle was still hot so it hissed and popped, looks like the only spores that didnt die of heat made it to the outside wall. Just made my first transfer 9 days after inoculation.  Its hard to see the culture in these pasty plates, but it looks like im finally making headway.    I did a transfer today, and I also dropped a drop of LC I made with WBS soak water and the same MS.

Low nutes does make it grow a little faster but thatโ€™s for myc transfers.

For spores high nute with soft agar is best.

I wouldnt even think you need to take out the old puck. Just pour over it it the culture was clean

Hmm... I did 10g Agar Agar and 8g Light Malt Extract per 500mL distilled water. Where does that fall into the high/low Nute/Agar category?

Sounds great for transfers, a bit stiff for germination. Its all about how much access the spores have to the nutrients and water. They cant get to it if it's locked away too mucj

Damn... I hope it works out. I nocced up 10 more plates with that recipe tonight. Using those ingredients, what ratio do you recommend for germination?

Listen I do 10g agar , 7.5 nutes , 500ml h20 and I get good germination rates from spores. You'll be fine

You got a ways to go on germ plates. They can take forever.

Thatโ€™s a fairly average recipe. Like he said youโ€™ll be ok, Iโ€™ve never had problems with plates like that either. The only time I had spores take forever was on plates with like 3g lme for 500ml.
