I poured and nocced plates for the first time last night. The plates that I nocced were still pretty liquefied and the nocc loop went inside of the agar instead of across the top. Assuming that the spores will germinate is this okay or will the mycelium not be able to breathe?

Ahh that's dope! I was considering getting the same type of storage cabinet for the worktable. Only thing that stopped me was how inefficient it'd be for the 100x15 plates vs the smaller ones.

I have an aa culture that I I thought was toast, there was mold all the way around. Now I am confident there is nice looking mycelium in there that I want a piece of, going to try a hot pour tomorrow, see if I can salvage a viable bit.

Thanks <3. Finally getting nice even growth after spinning my wheels on MEA. For some reason this culture goes tomentose on half strength malt but throws ropes on full strength potato dextrose.  :uhoh:

Yep, it's 3 transfers removed from a swab. Just cleaning it up. The original swab was a bacterial mess. Had to warm pour to get it clean.

Yep, it's 3 transfers removed from a swab. Just cleaning it up. The original swab was a bacterial mess. Had to warm pour to get it clean.

Bacteria can't handle as much heat as myc so when it gets meshed into a culture it helps to pour warm agar over the culture and letting it grow through it before you take your transfer.

Just don't do what I did with my first time trying warm pours which was not paying attention to the thickness of the original agar plate. Mindlessly pouring hot agar onto plates and boom, fucking overflow everywhere. SMH

Thats good to know. This is the first I've read about it. How long does it normally take for myc or bacteria and mold to start showing on plates?

A couple of days, but it depends on the culture. Requires some vigilance. It's mainly for bacteria-ridden plates, in my experience. Some swab plates that had competitor molds on them ended up beating the mycelium to the surface, in which case I had to start over. Myc seems to reliably beat bacteria, though.
