You certainly don't have to start back from spores unless the entire plate is coverred in mould.

Few new plates showing promising results.genetics are "Brazilian"
I'd say plate B/16/1 is ready for grains.
Plates are a standard 10g agar 10g LME nothing special, just distilled water.

Hey guys! Was curious to hear everyone’s opinion on how much of an agar plate y’all use for your master jars. I make my wbs jars around 3/4 full so I can expand it to 10 oat jars. How much do you recommend I use per jar? I make two masters per variety and am doing two varieties, so that’s four jars total.

I use half a plate split into multiple wedges. You can get away with less but the more nocc points, the quicker the colonisation time.
I only do 2 masters per variety also, i like to get them colonised quickly so nothing else has time to germinate. Once I have 2 solid masters I'm happy. Everything is g2g after that

Sounds gravy, I’ll make sure to use at least half a plate per master, may even go for 3/4 if I have enough good growth on these plates. Looking good so far.

No, not exactly. You'll likely get most of the phenotype, but not all because there are simply too many "branches" (for lack of better term) in the mycelium to get those narrowed down. Picture a pie graph, with thousands of tiny slivers each representing unique phenotypes. When you take your first clone, as you just did, that's cutting the pie into a single slice. Now that slice still has a couple hundred slivers, but far fewer than the whole pie. So you can continue to clone and narrow down the genetic pool, and that will get you closer to those exact genetics, but it'll take some time.

I first got into this hobby with MSS to brown rice.
Then I tried agar and wasn't 100% if I was doing it right, had some funny looking plates.

I didn't bother for a while as my plates never looked as good as the ones online so I stashed them all away.

I recently found the plates, many of them survived. Clones to transfers to 2nd transfers survived. I then gave it another go to take one of 2nd transfer and do a 3rd but got green nasties. Looks easily recoverable if peroxide will work. How effective is 3% solution? Pour onto plate or cotton bud?

Making a bigger SAB tomorrow.. Will definitely help.


Cookie rack or no rack? Towel or no towel? Bleach water or warm soap to settle air? I used a mini kitchen gas jet with the blue flames to sterilise my scalpel but not sure if it got hot enough. It got red but would fade away quick.

I just want to get this shit right. I've ordered dextrose too to try LC. Any tips or sterile technique tips would be great. Arm hole cover recommendations? Any order of spraying and not spraying? Thank you

So I did a bit of agar work this weekend. I decided to test some variations. FOR SCIENCE!

I did some glass(pyrex), some Celtreat and some Alien Holy Grails(pp5). I tried to use Cap's SAGARI method to see if I like soft liquifiable agar when shaking my grains. None of them are pre-pours. (Sorry Alien) Oh yeah, and I did some with a crap ton of food color and some with none.



So far I hate dropping pyrex on a tile floor with no shoes on. That sucked and lost me 1 & 1/2 dishes (saved one of the lids). The HGs and Pyrex came out of the PC a bit wet, so I let them sit in the oven at 200f for about 45 minutes before moving them to the SAB. After pouring Saturday evening, letting them sit in the SAB overnight and doing a few transfers Sunday, you can see that the HGs and Pyrex are clear and the Celtreats have condensation. I used a water bath to get the agar bottles to 45c before pouring, but maybe you need to go cooler for Cap's reduced agar recipe?

Anyhow, so far for ease I like pouring the HGs. Less condensation and no fucking around with saran wrap or tape. I set plenty more of each dish aside to watch for contamination. I guess we will see what happens.

My plates will be ready in less than a week. I was wondering what people think about using the edges of the agar plate. I used to cut around the edges and cut the center piece out before sectioning and dropping them in jars. People told me the edges can harbor contams. Anyone just use all their plate without cutting around the center or edges? Just want to make sure my masters are perfect.

Most of my plate contams are on the edge, soo it just feels safer and more natural to slice the myc out instead of transferring uncolonized agar.

Cookie rack, definately! As for the towel and bleach water, I've been running a dry SAB for about 4 months now with no issues.

I let the air settle for as long as it takes to smoke a spliff, iso my arms and gloves, sanitise my plates and crack on. I only clean the SAB maybe once a month

Don’t waste time with peroxide. Just practice transfers by . . . Making transfers. . . You can transfer off a dirty dish and have plenty of success. . . Just don’t transfer the contams. . .

Rack yes as gravity is your friend concentrating comtams on the bottom of your SAB.
Towel :shrug: personal preference, I’d suggest placing your rack higher rather than a towel. . .

Gravity to settle air. . . Before getting a hood, I left my SAB setup, and never sprayed walls or roof (thanks mushboy for confirming my madness that dry works fine). . .

Red = red, so is hot enough. . .

If ya can’t do agar, LC will make you abandon all hope. . . Practice agar. . . Then practice some more.

Any fabric will work as a cover for arm holes. . .

My tips:
Open petri as little as possible = work quickly through a ‘cracked’ dish, just enough to get the scalpel in and out. . .
Imagine a flow of dirty air travelling downward in your SAB, and keep things out of that flow where possible.
Wrap your petri from the moment they gel (solidify), and only unwrap when transferring. . . Then rewrap promptly. . .
Practice, practice & practice. . . It’ll come really quick. . .

I use clean quart ziplock bags and shove 3/4 in each inbetween the pour and inoculation. That way i only need to wrap once

Is Medley Hills Farm Malt and PowderForTexture Premium Agar Agar Powder any good?  Been using premixed mea powder but its almost gone so decided to mix my own up.  Not sure about links to products but all these are from amazon.  Here is a APE plate for your viewing pleasure.
