Hey gang,

Not much on asking questions, but I’m hell on search and reading. I’m learning that’s for sure. Had enough success that I’m still trying.

Anywho, a B+ clone transfer that I want to keep growing.
I read here about transfers, take what interests you.
I like that.

But this ones got me slightly stumped. I like it all.
Damn green dots....

So could use some advice for where to cut...

In a SAB, Id try for the areas furthest from the contam. around 12 o'clock area

Also I try to keep my transfer rice size or smaller, but I’ve always cut against the grain, so to speak. Can I cut length wise, like trying to isolate single strands of myc...

You can. People use punches, cuticle files, heart shaped tools ect.
I would just aim for the leading edge on that growth up top. The contam is likely from you, so just be sure to be aware of your hand placement when making the xfer. I would rotate that place 180 and have the section you are going to xfer from closest to you

careful with how you handle that plate as well. it looks like the mold is sporulating and you dont want to further compromise the cube myc

lol yeah not a coincidence there’s one in the exact same spot on each side.

So I just made a couple agar plate using a couple fat PE. With the agar plates will grow this exact isolated genetics?

Nah it should still work fine. I take the smallest sample I can with some tweezers. Usually about the same size as my agar transfers (grain of rice-ish).

Awesome,  I just cut out the pieces with a flamed exacto knife.

I’m all about small transfers too.

lol I love cloning huge stems like that, makes it so easy to get a clean piece.

I cloned one similar to those but shorter, it looked like a cupcake. Didn’t have a huge pinset but put out a lot of weight. Rock hard solid fruits all funky lookin.



That somehow ended up being about 400g wet.

They look awesome. :rockon: I just ordered this pack of petri dishes, Wondering what is the best way to make them. Make agar and sterilize it then pour, or pour them then PC the dishes?

You cannot PC polystyrene. If you want to pre-pour you need PP5 plastic. With those dishes you'll want to pour your sterile media in a SAB or in front of a flow hood 👍

Plates are expensive. . . Cheap financially, expensive environmentally. . . Polystyrene is one or the worst plastics also. . .

I prefer post-pour (Aka pour) with glass also, a bit more of a hassle, yet much less condensation. The sous vide has reduced this remarkably as pouring at 46°C has made a huge difference, though glass are more susceptible to temp changes

What genghis said, though make sure you pour the whole sleeve in one go. Once you open it, sterility is compromised. Pour them, wrap them, they will store for months :thumbsup:

If you have a plate that doesn't look great should you still take transfers or start back from spores? can you bring a bad transfer back from the dead or does it take starting over?
