I'd be willing to wager atleast 80% of the swabs I got originate from members here honestly. I bought most from the sub. Damn near all the vendors there are members here. The guy I used to use had super clean stuff but unfortunately he hasn't been vending or even active.

I Imagine bacteria is just part the game with swabs since you're making physical contact with the fruiting body. Not much you can really do about that is there?

Update on this plate that I don't even know what to do with: looks like strong rhizo.. but it got me wondering, what really is happening when all these colonies meet each other? Do they "know" each other? Do they interact? Do they respect each other? It's fascinating to witness, going to see where the plate goes even if I have to toss it in a river somewhere eventually.

Happened to pull out a stack of plates from early June and saw this little guy haha.. This was one of my first plates from an Amazon mss

Have y'all every experienced your agar losing it's strength over time?

I have some telephone brand agar purchased in 2018 that seems to be getting softer and softer every time I make up a batch. Last night I poured plates and they never solidified, just stayed runny.

Today I made up a bottle and had to use 15 grams of agar for a 500 ml batch of MEA to get it to properly solidify. This is the first time I've done MEA. In the past I've always used PDA.

I thought that my scale might be a little off but I tested it and everything checks out.

I have experienced this quite a bit but it's definitely anectodal. I went the opposite direction though, PDA from MEA originally. But I am still also shocked that 6 month old agar can germinate without contamination at all, so that's a good sign in general. But all the 6 month old plates did not seem to grow as strongly.

Yes that’s bacteria coming out from under the myc. You could still get a clean sample from that though.

Thanks, those are the only two with visible contams out of 16 transfers. Fairly happy about these results from syringes, looks like I have better luck than most and some of those syringes were 5+ years old.

Can anyone tell me what the invisible rings indicate? Most of the mycelium on my transfers look like they disappear for a cm or two before coming back. Does it indicate a bacterial presence?

The plate is a clone on LME @ 14g malt, 20g agar, w/ ampicillin.

Fuck no-pours. I finally caved and bought some celltreat plates. Pouring your own plates is legit less work than no-pour.

Yeah it’s super easy.

I was all stressed the first time and then two minutes later had a stack of plates cooling and was like ....that was it??

Make sure you check for cracked plates though they suck. I could not figure out what was happening the first time one in the middle of the stack started leaking all over.

I haven't had many problems with no-pours other than visibility and condensation, but I feel more confident with these plates already. Just streaked RW, KSSS and Melmac.

Yep, the visibility is much higher with polystyrene and the lower walls make it easier to make precise cuts and pick up wedges with the scalpel.

Its also super easy to clean up condensation on them. I like to put my poured dishes into a gasket box with some desiccant packets, inside slightly unzipped ziplock baggies, and the condensation goes away in a few days.

I do still prefer pasty plates for inoculating grain though.

Is there a name for this phenomenon? Whenever I search for stuff regarding rings it usually refers to the outside leading edge.
