Yeah that's what I was seeing and talking about also. It looks like that end isn't growing the same as the more uniform end. I'm not going to use that end, I'll use the other end to inoculate with when it's more grown out. I think the first two are the best because it's uniform throughout, plus I want to finally put something to grain.

I was thinking about cutting out the whole circle as one and putting each in a jar... Are you saying I could just use one of these cut into four pieces, and each quarter would be good enough for one jar?

I’m liking the looks of this one. Taking transfers from 2, 4, 7, and 10 o’clock.

I noc 10 jars with one 100mm plate, using 1" wedges, follow your heart.

@Staytrippy420, looks good, sounds good.

Found this plate thought it was weird that it didn't grow out so I put it aside. Noticed today it was stuck to the lid of the plate but still managed to get to the agar via tiny little threads of myc.

I've had my wedge get stuck on the lid but mine have never reached all the way down... that's badass!

How do these look? Tell me everything. First transfer from different germ plates from different spore prints. All Golden Teacher. Thinly poured 20g agar 10g LME 1 liter. Thank you in advance.

Four plates, top and bottom. (1,2,3,4)

All banging plates in my opinion but id transfer 2 a little more to get the perfect round.....

Is it okay to go to grain even if its just the first transfer? For the ones that look good?

You can got to grains and new plates.  Just leave a little on the plate when you A2G, or transfer b4

Here are these plates a few days later: I was going to knock up a few bags of grain tomorrow, but they started looking fluffy. #3 didn't get fluffy. What do you think now?

I reccon tomentose growth on high nute agar.  As said b4 try a jar and transfer too.  Why not?

do you consider this high nute? "Thinly poured 20g agar 10g LME 1 liter"

I personally do 14g LME and 20g agar (or less, depending) and get great rhizo growth so no I’d say that’s a low nutrient mix

Okay, so are 1,3,4 plates still looking good for grain? (clearly #2 looks funny) That rise in the recent growth looks funny to me.

Must be the culture then.  Personally id use 3.  But if you use the others do a transfer to new plates anyway.  You will tell if its good in the jar and uve lost nothing and gained experience.  That is a low nute formula.  I usually go 16g nutes to a litre and call that low.

some looking good ; but the others not so much

on your experience  , what makes the myc so fine and thin ? they looked really good before they were transfered , i got only good looking sectors etc , but then they did that

Standard MEA is 30g LME , 20g agar, 1L water

But for mycology purposes we call that "high nutes". Shroom standard is usually 15g LME, 20g agar, 1L h2o. technically that is half strength MEA

So at 10g LME per liter, that is low note even by myco standards, being 1/3 strength of standard MEA, where most people use half strength for general purposes. On slants I use full strength

Nothing wrong with it tho, it will work great. Will probably see faster lateral growth vs higher notes. Often more rhizo on low notes since it is spreading out to look for food

This culture came from a lone fruit of malabar in a triched out tub last fall.
The culture came out tomentose and provided more triched out pencil boxes when spawned. I had major issues last fall with contamination and buckled down on just agar work and small spawns in pencil boxes.

I switched to adding wine tannin to add more bacterial and fungal resistance. (being that cubes do colonize wood I figured I may be able to isolate it from other fungi and bacteria that cannot take a tannin laden environment that wood has.  Tannin is great in wine for keeping competitors at bay when making and holding wine for lond periods.)

First run was high nute 16g LME 10g agar 1/2 gram of wine tannon to 600ml.

Cultures were still looking sad with little change so I made another batch with 3g LME 10g agar and 1 gram of wine tannin.

I figured the high nutes were providing to much available energy to help the invaders overcome the obstacle so less nutes more tannin. I took one plate of the new mixture, opened it up and took a tour of my house with the furnace running, spinning around like a fool to see what would grow on it.

Results, Only one tiny spot had a trich growth after a whole week, normally it only takes 48 hours for the mean green from what I've seen and no bacteria or other molds whats so ever on the test plate.
That pretty amazing when just setting a normal MEA plate down for a session would create a jungle of contams and bacteria in 3 days were this one had a grand tour of unsterile fun. :takingnotes:

Out of 5 plates with 4 wedges with low nute and high tannin; only three cultures grew from the wedges and they were nice and ryzo instead of tormatose. Only the strong survive in nature right? :awesomenod:

Made some more plates with the first tannin recepie with high nute low tannin and did about 10 tranfers till the ryzo was less thick and suspect of ridealongs.
The cultures are looking great and performing almost 3 times faster.

Starting to look like some tricks from wine making can be reperposed for this hobby.

My hpothosis so far is tannin can be used as a natural antibiotic and fungal resistance additive and may possibly right out kill under performing or sickly cube mycilium.

Nice to see organizational growth in my sub again instead of sickly looking cotton. :magichat:

Well I knocked up my couple jars of the AA+!!! Whenever I was shaking to get the pieces in good placement in the jar, some mycelium rubbed off the agar pieces while shaking and jiggling around, and looked a lot more bare than when it was all rhizo when I put it in... Is that going to affect anything in any way? I’m worried because all the mycelium rubbed of the agar pieces when shaking and jiggling around.

R.I.P.Zappa what a fascinating observation... one might wonder if your tannin discovery could be applied to spawn/sub in mild enough quantities to help inhibit mold while allowing mycelial growth...
