Some more pics of my agar for some advice:

Here is the first petri dish. I thought this was mold but it has sprouted some good mycellium i think:

My guess would be to do a transfer from maybe 6 o clock or 9 o clock from this one

Petri 2:

For this one, maybe transfer 2oclock or maybe 7/8 o clock?

What do you guys think? does this look like something reasonably healthy or is that all just mold?

Doesn't help to show the top without putting some light on top.

I have a few agar plates that I transferred once already and now they are on second plate. No contams yet as it has been several days, but still quite small.
When is the best time to do another another transfer? Is it risky if its still small? About the size of a US dime. Too soon to transfer or is it too risky?

Hey guys. This would be mold, correct? All my plates of this specific strain are looking almost cottony. I used the same technique for my other dishes, a pe, and everything’s going smooth. Not sure if the condensation is causing this to fluff up. Thoughts? Any help is greatly appreciated!

There’s definitely cube myc in there.

Looks like you could get some clean transfers off

I would grab a piece of the leading edge from the first picture at the bottom sound 5:45 on a clock face. There is some good rhizomorphic myc there you can see the little ropey strands

Just want to thank this thread. I just made 9 germ plates of cubes... streaking the spores... analyzed the hell out of them with jewelers lenses attached to reading glasses and saw how dirty spores are. Out of those 9 plates, I saw three places I could grab clean mycelia. Now I have a bunch of plates growing out cleaner than I have seen in a while.

Many many thanks! I was too uncritical before.

dang bro , makes me skeptical of my plates ha  , how many xs on that lense ?

also , need you guys advice as of why the cultures that were looking very rhizo and organized, now look so different and not really good and strong? could they have contamed in the transfers ? the one's that look rhizo were petri dish clones , not the cultures that were looking good , it's like the mycelium didn't like the same weak in nutes formula , could that be it ?

one changed was i was using baby water , and switched to drinking water , could that affect it so much ?

could a standar LME formula stabilize the growth ?

thanks

Those plates look good to me, they’re just sectoring off nicely.

thanks man , but not so nice as the plate they came from , that ok ? i'm feeling like it's not liking it , or expected the same pattern of growth maybe ? anyway , i'll try a more common agar recipe to continue transferring and hopefully get nice and more stable growth

Those look pretty good to me, id take a transfer or 2 and then drop most of the rest into some spawn.

There's a couple of dots but that probably from premature inoculation ... lol.

They are some clip on jewelers flip down magnifiers. two of them. $5 at harbor freight. Really helpful.

Quick question for the guys that do a lot of agar work... what do you do with the plates after you take your transfers? I've got dozens that look kinda goofy so I dont want to waste the grain, but I also feel responsible for the organism I started growing and dont want to just toss the plates... is there an in between?

lol I saw somebody that cased plates and it worked recently.

I’ve been just throwing them into the box they came in. I recently dug through them and found tons of pins so those are good for cloning.

Or you can always put them somewhere in nature to go back to the earth. I used to bury all my stem butts and substrate. Kinda stopped towards the end of my last mycology runs.

I like this idea... my mother in law does a lot of gardening and composting... shes an old school hippy too so I can probably just have her compost it all... Thanks!

Are these good enough to go to grain? They are sort of splotchy lookin, which has me concerned.
