I gave up on my GT because they all seemed fluffy contam looking, all the way to T3s. So maybe I should have just knocked up a jar to see. :shrug:
You can see GT in my log, always fluffy, never rizo...



This is a good plate on agar with high nutes

Yes.

I didn't cause of the dogma.  Took T2 to grain.  Should have just gone with the T1.  But I'm still learning.

Then I have a different variety that is taking until T4 to be ready (except for one T3).

Once you know what to look for, go by the plate.

All banging plates in my opinion but id transfer 2 a little more to get the perfect round.....



Is it okay to go to grain even if its just the first transfer? For the ones that look good?



You can got to grains and new plates.  Just leave a little on the plate when you A2G, or transfer b4

This one seems to have a similar aerial fuzz in the middle as mine on the previous page, I’m guessing that is normal? If it were a contam I am guessing it would show black pin heads or change color before the mycelium reached the edge of the plate?

I love aerial fuzz.  Tilt the plate, it may actually be rhizo reaching for the heavens.

Contams grow faster as well...easy to spot based on their explosive growth compared to mush myc.

Thanks, I started using a cork borer for transfers and switched to LME then it was so easy that I did 25 transfers off one plate. I was gonna be bummed if they were all bunk. To grain some will go!

Looks you have plenty to work with, best would be 7, only looks like 1 large growth, 6 looks good too, just take a small transfer from outer edge... look at more pictures of good growth and compare :smile:

So I don't really know what I'm doing with agar. I am on my second mono with more grains colonizing but I don't know how I should be going about my agar work. Do I just keep isolating strains? I've taken 2 prints and plated one clone, as well as starting a new round of isolations plus my first set of isolations that I have been working on for about 6 months. I feel like I'm just doing what I see other people on here do without having an end goal in mind. Need some guidance because my refrigerator is slowly filling with plates...

What are you trying to achieve?

If you want to grow lots of shrooms, grow 1 MS grow, clone 3-4 mushrooms and test.  If its decent keep that culture, if not toss it.  Always freshen up your cultures and varieties and unless you are isolating for any scientific reason forget it.

Yeah plates will build up super fast. I was just kind of floating around doing a ton of random shit too until recently I decided to just focus on getting one good clone worth keeping for each variety, and some I keep a clean ms plate also.

Gives you an end goal and doesn’t take forever, plus it’s super nice to have something reliable on deck. I’ve been doing ms grows to stock prints and take clones, then just doing clones. At first I was just stoked to have a bunch of prints for different kinds, but now I’m working on leveling each kind up to a stock of prints and a good clone culture filed away.

And then there’s you know....the piles of shrooms that accumulate along the way.

I have a couple transfers of AA+, and each one the wedges fluff up and then proceed to grow out. Are you saying that fluffing is a sign of contamination or bacteria?

Not necessarily. It’s probably just the myc consuming the last bit of the wedge before it jumps off onto the new plate. Post a pic.

Well I did no pours, these are looking from the bottom of the jar, and they’re about the size of a dime now. I couldn’t get the best straight on pictures because the light doesn’t do the same since it’s rounded glass, I may try doing those in better sunlight. They’re both about a little less than dime size.

They both look good from what’s visible, especially the first one.

I just dug around and found this pic of aa+ t2 plate.



We picked the same color lol.

I can’t even fathom how frustrating this hobby would be without smartphone pics and this site.

Yeah, they look good from underneath, nice clean uniform growth. These are T2. I’m just going to let them grow out to full size and colonize was the whole agar puck, and then inoculate with those since they are clean. I’m not going to be taking all the way to 5 or 7 transfers. I’m eager to get this to grain, and it’s clean itself anyway, so there shouldn’t be any problems, and I’ll be inoculating with these two.
I’ll try to get some pictures in the sunlight today to show you what the actual culture looks like in the jar.

I just cannot settle for anything less than a perfectly clear view of my agar. It's sorta the whole point. No-pour glass petris only.

Pour from pot to plates on kitchen counter (at any temp)
Let plates cool, lids off.
Add lids, stack in groups of 5, wrap in foil.
PC for 20, let cool completely before moving off burner

Edit: also, I use jar rings and trivet, add just enough water to cover trivet.
