I can appreciate that, but there are faint, clear contams that can barely be seen through glass.

True. I’d like to think I can see them all but I most likely can not.

I always take back ups just incase.
Probably making more work for my self but sab work isn’t intimidating any more.
I actually look forward to it now

I’ve been having trouble gettting desired growth. Some details; roughly 1 week since Ms inoculation and this is what I have on every dish
Agar recipe: 10g agar, 10g Lme, 1g yeast
This is a bacteria growing correct?
What could I be doing wrong?
My procedure is pc for 30min at 15psi, I remove pressure regulator and let it return to 0psi. Then I remove agar and allow to cool until I can handle it without burning myself and let it almost start to solidify. I still get some condensation on the inside but not terrible. I Lysol my sab with a spray bottle then before I pour every dish I wipe both dish and lid down R-OH. After they solidify I begin inoculating by wiping lids with R-OH and using liquid ms to place a few drops on agar. I reseal and leave dishes in sab till I notice signs of life.
I’ve also attempted to let the dishes sit for 3 days and see if it’s a mistake I’ve made before inoculation but they remain clean.
I keep spores in a fridge that isn’t set to any extreme cold temp so i don’t think I ruptured the spores cells.
:takingnotes: On any advice this has happened with recently purchased spores and roughly 4-5 yr old spores.

Looks bacterial to me.
You shouldn't remove the pressure regulator, let it lose pressure on it's own. But since your control plates are clean, this isn't the cause.

You mentioned spraying Lysol, which isn't helping. How long do you wait after spraying? If you must spray (which you don't) try opening one of the control plates as if you were inoculating, then close it. My guess is the Lysol droplets are raining trapped bacteria onto your plates, which is why I quit spraying in the sab.

What's R-OH?

R-OH is the functional group for alcohol. And I’ll try not spraying and trying to keep condensation low on the inside

They're both ATL7 and yeah I'd agree the one on the right looks a little better. The weird things I always see on Tamp and ATL7 plates would be a big red flag on like a cube plate but for Tamps/ATL7 it's pretty normal.

These are some other pictures I tried to get in better lighting so you can see inside and straight on. These are what I’m going to use to inoculate to grain this weekend, just going to let them grow completely out, they should be done some day this weekend. They’re all T2. It’s good clean growth that I can inoculate with and put to grain don’t you think?

Your pics are improving. Your cultures are improving. Congrats.
I can't see much in the first 2 plates, they may be clean, need clearer pics.
That last pic is the clearest, is that yellow spot above your thumb on the surface? If so, it's bacteria.

No that was just the lighting and the curve of glass reflection. That plate only has good symmetrical growth and more rhizo on end, the other end doesn't seem to be doing that though really, even though it is still spreading,,, So you think the first 2 ones then are ready for grain? They'll be about the size of that whole bottom it grows fast on this agar. Should I cut out each one whole or cut to pieces?

If it's not bacteria in the third plate, I trust it most cuz the other pics aren't clear. But I'd let it grow out til about half an inch from the edge, then cut one inch wedges, one per jar. The bigger wedges colonize the grain a bit faster, your call.

They look pretty good. It might be the angle but the very right side of the last pic growth looks like it might be a little different.

They look good for t2 though, but I’d definitely recommend taking a tiny transfer before graining or leaving a tiny piece on the plate when you cut the myc out.

Once I get this culture cleaned up enough for grains I plan on making swabs. Shoot me a PM and I'll hook you up once I have some if you haven't already sourced one by then

Hello Shroomery,
I loveagar, it’s fast and efficient.  Thanks Bod and others for your TEKs, I don’t think I would have made the leap so quickly if it weren’t for you all.  I think I nailed a clone culture on my first attempt.  What do you all think?
