Magnesium will make the gel more elastic but I prefer Calcium Chloride which creates a brittle network. That's how I'm able to easily create a liquid inoculant with a stir bar.

Mind sharing the recipe? Also, do you use a different recipe for plates for cabins or plates  that's not going to be used for LI?

You can go with .2g or .3g of Calcium Chloride for cabins. 0.1 for slurry. 0.2g for an all arounder.

You can't just replace the chloride with your lactate though. Lactate is heavier

Yeah, so the elemental calcium concentration for chloride is 27% and 13% for lactate. Would it make sense to double the calcium amount if I'm using lactate?

My Calcium Chloride is anhydrous so it's 36% elemental calcium. You can multiply my measurements by 2.8

General question for anyone- When you use grain water to make plates how much grain water do you use for 500ml water & 10g agar agar? I usually use like 7.5g LME but wanted to try grain water if I am already producing it and there isn’t a difference.

Some Mexicana hybrid called "Planet 9" (ATL#7 x Mystery #2)
Showing signs of "tanning" on agar and also has some huge bumps on the germination plate which I think are stones forming? not sure but definitely keeping my eye on these plates.

Is this dot mold on my x7x+ or did I drop a piece of myc last time when transferring? I transferred away from it just in case. Otherwise these look pretty clean to me but how do they look to you?

Hard to tell from the photo if it's mold or bacteria, but it's a colony of something contaminating that plate. When I see a satellite like that appear so far away from the inoculant, I assume that it occurred when I was pouring the plates although it could occur anytime of course.

If you think you see a good leading edge 180° away from that satellite then take a wedge and put it on a clean plate now. Otherwise, it's far enough away that you probably have enough time to let your culture grow out a little bit so you can get a good wedge to transfer. The longer you wait however the higher risk you incur.

Looking closer, I think I see mycelium, so could be mold or fungus or it could be a satellite of your original culture if you weren't too careful about transferring it. How did you plate the original culture?

Poorly :lol: I started from spores and then tried to take some aerial mycelium to t-1. The satellite texture is different, looks like tv static. I doubt it's cube myc.

The yellow blob on the t-0 is interesting, I thought it was clumped malt extract but it seems to be growing.

I guess the take away is that something is wrong with my sterile work.

I’d be willing to bet it’s mycelium. This happens to me a lot kuz my tek isn’t perfect and I end up dragging it or dropping it in the plate I’m transferring to or have trouble getting it off the scalpel and that’s when it happens
