Malt extract arrived today, getting to work in a few hours. 10g agar agar, 10g malt extract, 500ml water is the basic recipe if I understood correctly?

I made a batch of agar the other day and saw a post by beef saying it has antifungal properties and I have noticed whispy basically invisible growth on some of the plates Iโ€™m imagining this could be why? Should I make a batch without any coloring or with blue and transfer everything? Did I kill the cultures or will they bounce back Iโ€™m sure the nutrients in the agar are better then the sterile media they were originally in or will they be weakened because of that? Typically donโ€™t like to get paranoid but if someone can ease my mind itโ€™s GREATLY appreciated

Idk personally though I've heard of using antibiotics in agar plates to cleanup dirty spores from bacteria to much antibiotics slows or stuns mycelium to little n risks bacteria building antibiotic resistance (I'd think a little red dye wouldn't hurt in very small amounts though this is a unbased opinion with no personal experience with it

When you guys do transfers do you put the transfer in the new agar mycelium facing down or up? Iโ€™ve been putting them downwards so far but Iโ€™m curious how you guys are all doing it

Up, but it doesn't matter. It's just the flow of movement I like when I'm doing transfers.

Is there any real benefit to doing more and more transfers?

For example, I have a few plates of mycellium that I don't plan on growing any time soon. Would it be beneficial to continue to do transfers?

Has anyone seen a difference in the way their mushrooms grow on transfer 2 vs transfer 20?

Unless you plan on growing out each new transfer I see no point in doing them, for most species I find that it's nice to try different cultures when you first start from spores, but after that I usually clone, grow out that clone, and take prints for backup and sharing.

Very hard to see but thereโ€™s some contam to the immediate right of this tidal wave, the only one out of all my pours to get contam so Iโ€™m glad to know it was my sterile procedure rather then my agar. Anyways should I try and transfer this away now to my new agar or wait and try to grab a piece after it grows more? I feel confident enough that my plates I made SHOULD be clean I did the same procedure and none of them contaminated except the one mentioned. Also I noticed on some of the plates the mycelium seems to grow โ€œintoโ€ the agar maybe a bit more then it is growing on top of it. Is this normal or am I maybe being deceived by the mycelium eating the dye? It could it possibly be the red dye itself?

I would place the cup on it's side in a position that would make the bacterial contamination slide downwards and give the mycelium a chance to grow upwards away from the contamination for a couple of days and the do a transfer.

The first few at atl#7, tampenesis. Really need these to work.

The other ones are Jack Frost, not as important but was really hopeful for these

Yep, they're plates alright. First few are kinda wispy. I'm presuming that's a thin layer of myc around all of them, and not some kind of film or other thing...


I'd pull anywhere from 9 to 12 on this, probably about 11:30.



Favorite of the bunch I think, bit faster looking and the thin perimeter is smaller. Thick bits at 10:30 and 1, maybe 8 just for variety.

What's the story on that lil intruder? If you dropped the wedge or otherwise know that's myc/expeected, I'd grain that (maybe chopping around the lil guy just to be safe).

But like.. what are your goals? If you're chasin ropes, T8 is a bit long, I'd start switching up temps (higher) and nutes (lower), but if you just want something stable enough to send to grain, you're there.

Yep, those are plates alright. Are the lil bits in the ATL bubbles/imperfections in the agar? Extra bits of myc? Not expected to be there? ATL is looking pretty even otherwise, I'd let em fill out and run it, as long as all those lil bits are benign. Very hard to tell anything in detail from the moto razr pics.


Is this a germination plate or is that two transfers? Hard to see but I'd take 1 or 8 from the top one, and 4:30 or 8 from the lower one.

The other one is kinda whatever, I'd probably take the right side of the thick part at 11:30 on the top one, and then whatever else on the perimiter just to have a backup. I guess 11:30 on the bottom part since it seems fastest. But all these JF bits seem early and like they could use another round or two.

Are your temps things that will make the tamps happy? I can see your temp cycles in the ATL ring density. Not a bad thing necessarily, just wondering.

These were on the same day, same plate. Left is mashed potatoes + Karo, right LME 2%.

I want to transfer from the PDA, or wait a few days and send it. The LME just seems weak, and I really want to do LME but I never get growth like with potatoes :donut:.

How do I get beautiful growth on malt?
