Some of the leading mycologists in the US are self taught and didn't go to school for biology.  But there are a few folks here who are pursuing degrees.  I'm just a nerd who likes to read biology texts and papers, so my understanding of some of this is sorely lacking.  But I do my best and I'm open to correction.

The following is my understanding and it could be wrong.

@kloakk two monos clamping is a dikaryon.  And they may be vegetatively compatible but not good mating partners.  In which case they might find a better nuclei to clamp with later in the lifecycle. 

They're also amorous prior to making mushrooms, which is the parasexual cycle I mentioned above.  Because two monos might not be good mating partners, it's generally easier to clone a good fruiter from a plate that has had more opportunities to find a mate, than to run out a bunch of mono/mono dikaryons.

To isolate a single strain, take small transfers from leading organized growth.  Larger wedges and disorganized growth are more likely to have multiple strains.  Using grab and drags or other germination techniques makes this easier to do with minimal transfers.  And can even be used to pull monos (which really isn't hard, but a microscope helps to confirm).

Variety is the preferred name over strain for a large set of DNA like Penis Envy or B+.  Strains are active dikaryons on a plate, in a jar, etc.

I'm mostly talking about semantics here, like the definition of a word has to mean the same for everyone if we are to understand each other. So 2 monos fusing makes a dikaryon, so if 3 or more monos fuse it should have a different name or everything just becomes muddy.

Like I said, I don't remember where I read it, it was on the org. Point being that a strain is 2 monos fusing making a dikaryon and that dikaryon is a strain. I have no clue of this is true or not, but naming different things the same sounds confusing and is why I asked.

And when you say find a better nucleic to clamp with, I am talking about putting 2 spores to a plate, they fuse and become dikaryon. Would there be any other nucleic to clamp with on that plate?

Would that even be possible? It would seem that would make for a competing nuclei situation and one would lose out?

Also, is anyone here incubating their plates or do you just let them sit at room temp? Any cheap devices to incubate ?

Don't toss anything until you have a successful transfer. Take a transfer from the consistent part to a new plate. Keep doing this until the whole plate is uniform.

Don't incubate your plates, room temp like your fruiting chambers is fine. When my dehydrator is on in my closet it warms everything up a couple degrees but there's no need to waste electricity on incubation.

Also you don't have to have a perfect plate to have a great grow, just needs to be clean and strong. You can cut wedges from some of the plates you posted earlier and inoculate jars with them if you want to

Dikaryon means two separate nuclei in a single cell.  Monokaryon means one nuclei.

Fusion doesn't typically take place until spores are made.  And during vegetarian vegetation, a dikaryon may meet a mono or another dikaryon that is more compatible for mating.  But the result of the cross is still a two nuclei dikaryon.

The process by which this happens I don't understand well.

I think I understand :lol: So at the plasmogamy stage the cells will fuse, but the nuclei won't, bringing two haploid nuclei close together in the same cell, using a clamp right?

And fusion of the nuclei happens in the gills before the spores are dropped, right?

So then mono + mono = dikaryon if clamps are formed. Dikaryon is what I assumed was a strain. When one or more monos or dikaryons mate with that dikaryon it is a multi strain colony.

Isolating is one thing(redusing the number of dikaryon strains from a multi-strain colony), but an isolate(one single dikaryon) I imagine is harder to do with just our eyes and a scalpel or?

Mostly right.  Clamps are specialized structures that help get one of each daughter nuclei into a new cell.  They indicate a dikaryon strain, but aren't part of the initial two monokaryon hook up.

Generally yes.  Parasexuality is the other time this happens.  But I'm realizing as I read more this morning that this hasn't been well studied in mushroom producing fungi.

No the blue is a multi-strain colony because I can see three distinct sectors and I'd be taking a large wedge to use them all.  If I just pulled the cross I would assume it's a single dikaryon strain.  It being the product of two dikaryons could just be the other two colonies donating a nucleu to make a new single strain.

I think if it's not sectoring and the growth is very organized after two transfers, it's likely a single dikaryon.  From my observations and what I've read, they stake out territory or hook up.  But they don't really share space or resources with their siblings.  I know that's not how we generally view things on the boards though and if there's academic papers showing otherwise I would love to read them.

Shakti coming along nicely, this plate has good examples of both types of growth, figure did share with the new guys.

I decided to take a transfer from the strongest white rope bc why not.

The cut looks janky because I have been lazy and just using disposable needle tips to take transfers unless I need to whip out the scalpel and flame it and slice. Saves me a lot of time and makes me more willing to bust out the SAB and make transfers when I have a few mins.







Alson while I'm here I'm wondering if anyone has used dyes that the myc didn't like, and how it behaved. It's not every culture on every plate but this aerial mycelial growth happens far more frequently on these red plates and I'm wondering if it's just coincidence. This is a Melmac plate doing it here:
