Hey,

Are the following plates looking good to you?
Sorry for the bad quality, I tried my best.
For me they look good at regular light, the only concern I have is when I use a flash light from the bottom, I can see some darker spots. Is this normal?

Suppose there's a big fat mono sitting there and on one side of it another mono slides in and that is dikaryon clamp history. But what if at the same time another swinger, from another mother, monokaryon jumps in and clamps up too on the other side? More interestingly what happens when the duality of the relationship meets in the middle?

Is this close to your B + E example from a page or two back?

This is the trikaryon? Double clamp connections possible?

Am I understanding this correctly as a real possibility(but not likely)?

So I suggest ignoring the b+e thing.  Parasexual cycles are common in other kinds of fungi, but I have not found any papers really talking about it in basidiomycetes.  So I feel like I was making a stretch considering other mechanisms can explain late crosses like dikaryons not being completely stable.  Which means they can revert to monos and that opens up breeding.

And I saw kloakk already quoted me :lol:

This was my understanding a few days ago. It could be wrong.  But I've started collecting some of the papers I'm cribbing from here.  And if anyone else would like to participate in the conversation to I would love that :smile:

https://www.shroomery.org/forums/showflat.php/Number/29551228

One I'm reading now mentions a three-nuclei fusion hyphae in di-mon crosses that links the two colonies.  Maybe something similar happens if a mono finds two mates in a timely manner.

It looks like there is a fusion cell with three nuclei in your and kloakk's scenario.

Quote:
It seems that a fusion cell in a Buller mating, having three different nuclei, has a different outcome than the pairing of two heterokaryons that have a common nucleus, also having three different nuclei. Thus, the difference between a Buller mating and a “common nucleus” heterokaryon pairing cannot be due to genetics, as both involve three nuclear types in the fusion cell. Instead, perhaps the success of Buller matings relates to nuclear migration, which does not occur in pairing between heterokaryons with a common nucleus

I think it's likely that these fusion cells are what is happening here in this late cross.  They avoided each other until one of the dikaryons broke down into a mono, hooked up with a neighbor, and bridge mycelium are why we don't see avoidance going forward between the three colonies.

How are these looking? Inoculated from liquid culture syringe on 4/17 with golden teachers and albino Avery’s. Realize that’s not the ideal method but I have a bunch of liquid cultures that have been passed to me…Have a feeling something is off. Was going to cut out the white and transfer and try the cabin sequestering…agar was 10 g agar 10 g lme 500 ml distilled water

I need definetly improve my agar technique..not always have nice results!

1 - I use no pour agar tek (the containers go to PC with mix);
2 - Containers that I use are that in foto - PP5 - and I can live the lid a litle open, to air change;
3 - Last recipe that I use is based in PGT123 but modified: I use 300ml of water, 4gr potato powder, 4gr agar.agar and 2gr honey (if I use the ratio 100ml water, 1gr honey, 2gr agar and 3gr potato powder the results are worst!) - with 300ml I made 20 agars.

Maybe the problem is with potato powder because have industrial ingredients and no pure dextrose?

The example in foto:
A - its a mushroom GT clone: and dificult to grow! Never can grow clones in this agars!
B - its a grain of colonized brow rice GT that take of spawn colonized and in this case grow.

Better use water from boiled potatoes instead of potato powder? Or definitly go to Light Malt Extract? I cant find in lical stores - and in Anazon only found Malt Extract (no Light) - its ok for agars?

Need definitly improve this..✌️

When you shoot a bunch of liquid onto the plates they tend to look like that. Something I learned from a fellow shroomerite is that if you squirt a bit of fluid from the syringe until you have a drop hanging from the end you can squeeze the walls of the syringe to get a single drop to fall onto the plate. Then you can pick the best growth for your first transfer.

Nothing looks crazy, just super dense and chaotic. Pick a juicy spot and take a transfer c:

You can find LME on Amazon.

And yes, if you’re struggling with agar I would recommend dropping the potato part until you get it figured out IMHO.

Stick to a basic LME/agar/distilled recipe then go from there.

Once I was more confident in my overall agar skill I went to an active charcoal/lme/yeast/agar mix.

As for pucks. Take transfers from B and proceed. 3 has some really nice spots.

I would grab multiple from the 2-4 range personally

In my experience, soft plates suck for almost everything (they're maybe a little better when trying to germinate spores). Stick to 2% agar (that would be 6 g for your 300 g of water) until you get consistent results and want to see how softer plates behave.

But, it's not quite clear what problems you are having. Can you elaborate?

The grain plate looks good from here, and the clone might be ok after a transfer or two, might be a little bacterial - stiffer agar will help you find clean growth, since it's less hospitable to bacteria. So are BRF pucks if you're having trouble getting clean culture from an agar clone.

Any malt extract will do - start with 1.5% or 2% of the water weight. Lighter malt makes clearer plates, which I don't think really matters all that much. For that matter, you can use brown rice flour, though you will probably want some food dye for better contrast. I've never used potato flakes for agar plates, but lots of people do (usually with dextrose though - there might be a conversion factor to replace it with honey).
