Agree, instant pot here too, great value and use for agar, grain, lc prep. just need a bit longer compared to a true PC. when I get contam is sloppy SAB technique, not the instapot.

Both pics in sig come from all instawork. Just need more time.

Edit. It’s Pudins like Al uses. While he says he wouldn’t recommend it, if someone has an Instapot and not a pressure cooker I would say start learning in the insta before buying a pressure cooker.

If someone has a insta and absolutely wants a PC, I sure as hell wouldn’t shame them. But RR once called cubensis the “trash can grows” so if a insta works, I have no reason to buy a PC.

It depends on what you are growing.  Cubes you can tell pretty easy when they make rhizomorphic growth.

Pans I look for leading hyphae.  Or mold spores.  :shrug:

Three years working with pans and this mold still fooled me.  But it turned green two days later.



But if these ones had been mold, they already would have made a mess.

Does this Charlie Brown plate look like it's exhibiting normal sectoring due to multiple genetics in one plate, or does it look contaminated? Two areas have me wondering. The area at 12:00 looks to me like sectoring because it's growing right out of the wedge. The area at 8:00 seems to have a dropout in it but nothing really shows up on the backlit photo. My gut tells me this plate is fine. What do y'all think?

I have a CB plate that looks similar and it seems fine to me too.

You could send it and hope for the best, but if you aren’t in a hurry, maybe make another transfer and see how it goes?

I ended up taking a transfer from mine just to be safe.

Recently streaked some mexicana and tampanensis, wouldn't mind a second or third or fourth opinion, been out of the game for a whfile, trying to get my sea legs back.

4-5 days for germination on these guys, seems pretty quick, but matches my swipe/streak patterns-ish, relatively uniform, lil bit of 'puffy' growth though. So often tough to tell early growth with the mex/tamp/copes etc vs. cubes.

Have 10 other plates that have very little to no growth just yet, visually, contaminate or otherwise.

Shitty focus, I apologize.

I was just reading about and thinking about starting some lion's mane.  Can it be cultured on agar only, and then moved to grain before spawning to a wood-based bulk substrate?  I'd love to know more about the agar with dowels method....

I took clone biopsies from grocery store fruits.
I did those on regular LMEA plates.
I’ve been doing no-pour agar in glass bottles for a while, and decided to add some 24hr water-soaked dowels.
I did transfers from the original clone plate, and put some of the Lion’s Mane in the dowel bottle.  These are  just for decoration, and I wanted to see how it would colonize given the added textural differences.

Also! Finally poured some 150mm Falcon plates.
Used some 50mm to take up the excess.
The biggins are real agar hogs.

Has anyone tried adjusting the ph of agar to 8-9 to combat bacteria and if so, what were the results?

I did 9 from a sleeve of 10. It didn’t feel like I had sufficient agar remaining, so I decided I’ll use the last for  some sort of big spore print this foraging season.

Just to train my eye. It looks like the top left and top right pieces has mated in the sector at 12'O clock. Or possibly two incompatible strains in the top left piece. Top and bottom ones have are not compatible on either side. And similar situation on the bottom side as in the top?

I think if I was gonna take one transfer from this plate it would be at 10-11 ish. My reason is that the growth is quite vigorous and I can clearly tell that the growth is regular and radial.

Please let me know what your thoughts on this plate is, if you have the time treefrog :lol:

The strains at 10 and 2 seemed compatible, and possibly made a new strain at 12.  The strains at 4 and 8 also seemed compatible, and possibly made a new strain at 6.

So, 12 and 6 each had a small transfer taken to a fresh plate.  As these are more likely to be strains that fruit well.

I also pulled three large wedges (10, 4, and I don't remember the third), because I had three AIO jars already prepped, and wasn't planning to use them otherwise.

I did a sleeve of grain water agar a few weeks ago, pulled them out to do some transfers and spotted one (from the side) that looked interesting. It's actually really interesting! Clearly picked something up, but I really like how symmetrically the contam is divided. It was the last plate of the pour (can see it didn't quite fill the plate), probably was lazy and did something dumb. Off to the trash, cool contammed plate!
