I want to know if there's any difference between placing the piece of agar with mycelium facing up or down. Does it expand faster or slower? Is there any significant difference? Because it faces downwards, does expansion begin earlier due to direct contact with the agar?

You asked this in a separate thread too. I think you misunderstand how agar works. The mycelium grows through the agar, not on it. What you're seeing on the surface is aerial mycelium, but there's also myc throughout the colonized part. It will grow at the same speed on both sides.

I, as well, thought agar worked because the mycelium grows on the surface, not through it.  which is why other thickening agents instead of agar (such as gelatin) don't work as well... or maybe you're right and it's just important that we can focus on the surface and observe and isolate, etc.

but in reference to the question: no matter the mechanisms, it doesn't matter how a transfer is placed on a new agar dish - anything works

Yeah in most 'agar 101' type texts, the suggestion arises that it grows 'on' agar, but the 2D effect we're looking for is created by the fact we pour thin. But well. 'An agar plate is a 2D workspace' sounds a lot better than 'An agar plate is a 3D medium with a lot of space over 2 axes and very little space over one axis, so functionally it's 2D'.

Adding a bit of detail (maybe too much, sorry):

You'd get the same basic appearance (radial growth on the surface) even with reeeeally thick agar. Maybe it would grow a little slower because it doesn't need to search out more nutrients. Any surface is inherently two dimensional, even if it's not actually a plane, like the interface between grains and a jar or bag, or the surface of a wood chip - and you see the same growth patterns there too.

It's definitely growing in the third dimension too (into the plate or grains or whatever), but the surface interface prevents it from growing "normal" to (outward from) the surface (other than a thin layer of vertical/normal-reaching mycelium), and so it grows parallel to it, covering the surface itself, radially.

I'm not sure it grows at the same speed in all directions though - in a solid medium it has to work a bit more to cut through, and I suspect there are practical advantages to e.g. enclosing a grain before completely embedding through it.

And of course we only really see what's on the surface since we can't see through the mycelium, and mycelium exposed to air seems to always be a lot whiter - in liquid culture, e.g., it's pretty translucent.

Which is more detail than we need for cultivation, of course. :smile: It's only really important that the agar has mycelium both on the surface and underneath.

I've noticed some cultures (oysters especially) make the agar a lot harder to remove in large intact wedges, and I assume it's because of how relatively aggressive they are (by the time I get around to cutting or cleaning the plate) at colonizing the third dimension of the plate.

A lot of the 2D growth on agar comes from bacterial microbiology. Bacteria will pretty much stay on the surface. Mycelium can grow through the agar, which is why cabin sequestering works so well.

I only send T2s+. Works well. And, yes, the T2s are generally more organized.

Also, you can pick how many T2s you want. I make 3-5x more T2s. Any extra plates go into the fridge for a while.

But, whatever. One system or another.
