No it's agar. LME is just he nutrient.

They're getting sequencing for a second time at the moment... I'm pretty sure. Or it's already been done and I've missed that part. Microscopy done by university in Costa Rica says moseri tho. You can look at the inaturalist observation by punching the numbers in

I decided to toss this plate out completely without taking a transfer. It looks contaminated to me and I think the contamination is too widespread. Looking back at the photo though I wonder if maybe this is just a combination of tomentose and rhizomorphic growth so, for science, I decided to post and see what you guys would have done.

Interesting. Officially it hasn't been tossed yet but it's in the toss pile at the moment.

I've cabin sequestered but I have never trenched. How would you trench that plate? Meaning where would you cut the trench?

Also, I would not be considering tossing it where it not for the fact that I have T1 and T2 plates like this that are looking much much cleaner and I've got them coming out of my ears at the moment

Yah I understand I got 3 decent plates of Tresure Coast I am just going to watch pin. Only because I have this much Tresure Coast all LC I just tested it on agar then did transfers to see what it looks like without LC splashing it around.

A trench is just cutting out a rectangular section of agar for the mycelium to reach across. It works the same as a cabin. You do it just past the leading edge. Only crappy part about that plate in particular is there really isn't much room to trench anymore considering the good growth is on the sides. So you would have transfer first. And of that's the case, my as well cabin it. I stopped putting roofs on my cabin. Screw all that extra work haha.

Here's an example of bottom and top view of same plate. I just face plant a wedge on the plastic and it grows across. I'm convinced that the roof of a cabin is really an unnecessary step.

Nice. Yes I have done CS with both a roof and without and in my limited experience found that it worked with equivalent success.

It always hurts me a little bit to throw plates out unless I've taken a bunch of wedges off of them first. They're like my babies, so I will keep this one and at least take some transfers and cs for grins

Don't throw the plates away. Put them on a shelf somewhere and come back for them for pins!

If you get pins on a plate, what's the best practice of what to do with them at that point?

Pluck and transfer to a new plate, free clone. :smile:

Sterilized tweezers and a scalpel help to get as little surrounding mycelium as possible with it, so that the pin itself will send out new mycelium.

Edit to add that for clone cultures (like the SGT), it's probably less useful to take a plate pin from. Clone of a clone at best might narrow genetics down a little bit more, but you don't want too many generations of that.

You have a ton of SGT mycelium now, no need to be precious about one plate. :mushroom2: But if I had a spare jar, I'd have no problem sending a wedge from part of that plate that looks rhizomorphic like the rest of the ones you've pictured.

Has anyone experimented with adding anti-bacterial compounds that might not damage mycelium in their agar recipes? I don't meant antibiotic shit, but like, black pepper, red pepper, tumeric?

Of those, apparently only black pepper would be harmless to mycelium, but effective against bacteria. Perhaps the others can still prove to be useful. Perhaps a way of training the mycelium so that only the most resilient ones survive?

Also, my gf made a beet salad. Next batch I'm coloring my agar with beet juice.

Beets are anti fungal don't bother

what does everyone think of my chestnut mushroom clone? I think it has mild bacteria and needs T2 but looking salvageable.

I grew them with masters mix but was thinking about trying CVG. I suspect I can fruit most mushrooms in CVG by just casing grain.

also i made some advances in the way i send wedges. These foam inserts are cheap and they protect the package from cold (maybe?) But also make it harder to 'feel' the ridges of the baggie.  It just feels right. also

I save bubble wrap from packages I get and cut it to size and it works pretty good too

I’ve started experimenting with cinnamon and green tea in agar recipes. Despite them supposedly being anti-fungal as well as antibacterial the mycelium is growing on the plates. Growth is different looking and it takes a few extra days to jump off the wedge, but it seems to be doing well. This is for stone producing mycelium, haven’t tried cubes.
