Low nutrient agar can be used for the same affect, the mycelium should run away from the bacteria faster(should) but there’s better ways to de contaminate than just using low nutrient agar.

With added nutrients it’s definitely growing you may just not be able to see it with the naked eye. You’d be surprised how far past the “halo” you can find strands of mycelium.  3 days is barely time to let the culture establish after being cut, let alone to let it venture out. Give it a week.



Reassuring.  I was very careful moving the slices, only cracking the plates enough to set the slice, and making small slices.  18 plates and only one with contamination, and it's all over.  No idea how that happened. 

Using a SAB but armholes open.  Today I sprayed the whole inside with hydrogen peroxide 3%.  After prior experiences I'm worried about this SAB.  I washed it all down with a hose a week or so ago because of prior contaminations.

I took several slices with plins and I can't tell whether they have contamination or it's just the aborted caps yet.  All plates sealed with clingwrap cut to width -- cheap and easy.  TEAM CLINGWRAP!

Looks like I'm on the right track with this 1% agar.  Maybe this will be the time I can go to grain.

Ok now I've developed one or two green spots on about 4 of my 1% plates.  Does no one talk about this, or am I alone?

Would it be a good idea to crack them and excise the green before it sporullates, to save the remaining cultures?  I have four quadrants each plate.

Worried that my SAB is just contaminated.  I could hold the plates vertical as I open them to cut the green.  Needless to say, flame-sterilized scalpel.

Oh contamination happens. Spores fly in through your arm holes or fall off of you. It’s a thing. Use 70% iso not peroxide. Let the box settle a little longer next time maybe. It’s a learning curve you really just gotta do abunch of plates and learn that way. Anyway that’s how I did it. Separate anything you care about from contamination asap.

Slow, mindful movements. Dont disturb the air or put anything over the plate that isnt sterile. Open things slowly. The air is full of mold spores. Use gravity to your advantage. You want gravity to be the prevailing force, not the air your hands displace.

Don't disturb the green , remove the healthy parts

I have so many plates that sooner or later something's going to get kicked up.  Thinking about doing bits on different days.

I've used alcohol and agree it is better.  But Supa uses H2O2 so I switched to that.  I am tempted to go back to alcohol, although it breaks down spayers, and I can only imagine being immersed in alcohol fumes and firing up that lighter...  FOOM!

I have always been afraid of fire, to the extent that my house (structure) is non-combustible.

This happens to be a spore portion of the spore->clone->spore->clone process.

Transferred a plate pin then noticed the gremlin under the agar. The plate was poured a month ago so I imagine it will probably stay down under the agar and give me time to transfer away.

If it works for you that's cool. But I definitely recommend making your own if your having any issues at all with your sab work.

Wow.  They're actually doing what they're supposed to!  Beautiful. 
Three plates of each strain,to hedge my bets. T3, T4.

       

This MiB pheno is the most ferocious of them all.  One of only two I've gotten plins on. 

And the 1% MEA is a winner, so clear and it works.  I was going to do charcoal but forgot at the crucial moment.  Maybe lucky.  Wondering about calcium?

Decided to cut out the green, with the plates vertical.  Not digging the rhizo growth, so selecting for this.  Results remain to be seen. (straight and tall, potent)  I've made my bed... now I have to lie in it.

Got my grain hydrated (boiled for 2 hours) and laid out to dry to the correct moisture, credit: BeefSupremeJr.

I've taken a page from ReverandMyc and am keeping a running log of every plate. (with a few mods)  Blessings upon you good Sirs.

I've had so much problem with contamination despite all my efforts (above), that I am hedging my bets.  Also it gives more opportunity to select for different types of growth.  Four plates in one.  I take a pic of each plate and put it in my spreadsheet to its numbered entry.

Also I can hold the plate in one hand, slide the lid over just enough to set the slice, then slide it closed with my thumb reducing contamination.  I am now holding plates vertical for most opening operations.

I can send a quadrant to each quart jar and track the results.  Gotta send the MiB soon now.

Lab (bedroom) is right next to the bathroom which can't be helped, but I'm keeping the door closed.

Curious about this as well…
I do 3, but also placing them closer to the center so they have room to grow out before reaching the potentially contaminated edge.
