Tc question.

I've been playing almost exclusively with pf tek for the last year.  I'd like to do more with agar and brf pucks.  The issue is my sab. 

I got a pretty good sized tote for my sab.  This means I have to put it away in a fairly out of the way place between uses.  It can only fit in pretty open parts of my home when being used and my family situation is such that it severely limits the frequency I cam get it out. 

What is the smallest tote you could use for some sab work?  I don't see using quart jars often.  I use a very small amount of mushrooms myself and don't have a circle that promotes sharing.  I want to do small grows and work on very clean prints, hence brf pucks.

Your advice is greatly appreciated.

Smallest I would go before feeling extremely squiggly about it is a 54 qt clear sterilite tub.

BRF pucks are good for germinating stubborn spores and cleaning up cultures, but they are not a substitute for agar.  Make sure to focus on agar if you are going to be going to grain.

Is there a reason you're trying to expand to agar use if PF TEK is supplying your current needs?  Agar means more supplies taking up more space and there's consequently more supplies to be found.

I've been dealing with increased contamination in my pf tek lately. I'd like to work with cleaner materials. 

Also, I've been flame sterilizing my homemade spore syringes but not working in a sab when innoculating pf tek jars. I'm sure easier sab access will improve it. 

Lastly (for this list) while I don't have much time for tripping the act of growing the mushrooms, knowing what they can do, is also somewhat soothing.  Nurturing the growth appeals to me. I've done a couple trades with members here but not contributed spores.  I feel like sending out some close-as-possible to sterile swabs or prints from brf pucks would be cool.

Enjoying working with fungi is a great reason to expand to agar.  A sab will absolutely help cut down on contamination.  My concern is the keeping it all secret from your family.  My hope is that you mean hiding it from your children, not hiding it from your spouse/partner or parents if you live with them.  The first is tricky but kind of standard, the last two are unethical.

When I lived in a studio apartment I would flip over my large still air box and store all of my myco supplies in it while not in use.  With the lid on and tub turned so the arm holes were facing the wall, all it looked like was a tub full of stuff.

So let’s say you take up agar.  Now you need a pressure canner for agar and grain, you need plates, you need space to keep colonizing grain jars, you need new fruiting chambers of whatever style to fruit in, you need more storage space for increased production.  Upgrading is fun, but it’s going to present you with new logistical problems.

Also, prints aren’t sterile.  They’re not even sanitary.  Anyone receiving a print should know this and know that it is their job to clean it up.

It's a kid thing.

For the "sterile" print ( I know they won't be) they would be grown on pucks in the jars.  I think that's as close to sterile as they can get.  This is from revmyc's tek. 

I know it can get dodgy modifying tek, but I'm thinking of maybe taking a flat syringe to take agar samples and use them to inoculate pf tek for the reasons you mentioned about space.  A combo of a simple lc tek I've read here (pastywhite maybe?) And pf tek. 

Also, on the other list of things I didn't mention in my last list, I often procrastinate making my spore syringes because of hastle and logistics of getting the sab out. 

I'm open to other suggestions on cleaning up my pf tek.  I don't want to come across as the newb who refuses to listen to tc's. I've done a number of pf tek runs successfully, both as pf cakes and expanded to coir.  I know good technique is almost certainly to blame here, just not sure exactly what to do to improve.

I recognize this is starting to ramble, nut realized while typing this that I think my score prints were taken before the mushrooms were totally ready.  I wonder if higher concentration of spores might help out compete nasties 🤷‍♂️

I think before you go nuts with trying fancy new things with agar you should stick to the basics of transferring with a scalpel.  I have made a number of PF cakes by switching out the four hole lids with some plastic lids that have synthetic filter disks on them, then transferring wedges with a scalpel in a SAB.  That's them in the bottom left corner:


Once you're working with agar sterility of prints (which again is not possible) is a moot point.  Agar is where cultures are cleaned, it is the bottleneck.

Ultimately the answer is this:

You need to at minimum be using a properly sized and made still air box.  Nothing less will do as your next step.  It's not negotiable.

Thank you.  I will look at going the scalpel route.  That seems to have no significant downside over the syringe.  Either would clearly need to be done in a still air box.  I'll take a look at the 54 quart size.  I recognize the needs of the sab, I just need a size that fits my life.

I don't see a big difference between the space an 80 quart and a 54 quart would take up, but the workspace and cleanliness would be a significant difference. 

There's a lot of downfalls to using syringes.  They are a contamination risk in several ways that a scalpel is not.  The reason it's better to go scalpel is there's less contamination vectors to consider when something goes wrong, and troubleshooting is half the hobby.  Being able to narrow down where something is going wrong and having the experience to see it is very important, that's why starting with the basics of agar and working your way up is so useful.  Think of it as a skill tree, gotta start at the trunk.

Can any TCs tell me what might be happening here?

Details:
Honestly I was just playing around with the shoebox tek with some spare mycelium (crumbled bits of PF cake from physical trauma, AKA my clumsiness, post dunking) and left over substrate I otherwise used for a monotub. The spawn was made with "Cambodian gold" spores.

The ratio of spawn to substrate was silly, like one part spawn to 8 parts substrate. But I was just curious as to what would happen and more focused on my monotub. My monotub produced more than what I reasonably needed, so I wasn't worried about the shoebox and mostly forgot about it. I would occasionally mist if I noticed a lack of evidence of humidity, but this was only every other week or even less frequently.

It's very cool in my house where the shoebox is, like 64 to 68 degrees F range in an average day. It gets some indirect light from a window on the other side of the room, but I didn't have my mushroom light on it at all. I wasn't in any hurry. When I would mist, I also looked for signs on contamination but didn't notice any. It also doesn't smell off, smells the same as my monotub did.

The shoebox has been chilling, literally, since mid March. Myc growth is apparent but very slow, but I figured the cold was causing that and again, I was just playing around to see what would come of it, if anything.

Today I did a double take as a noticed a tan circular shape through the condensation on the lid.... One lonely mushroom, veil already broken, just sitting there...

How? Why?

My only theory regarding timing of this is that the room may have gotten as warm as 74 during one or two days over the last couple weeks due to unseasonably hot weather days and it is getting longer exposure to light from the window with the longer days of summer. But that still doesn't explain the single guy/gui.

Questions: What is probably going on here? Any recommendations for next steps? Like should I try to keep it warmer and give it 12/12 full spectrum lighting, or is it unlikely to ever fully colonize or fruit so I should just continue with it as is just to see what happens?

Also the stem had a brown/reddish spot in the middle when I cut it, the other fruits (from monotub) had hollow stems or a uniform color in the smaller fruits. Does that mean it's contaminated/unsafe?

Cubes are tough, they will find a way to pop spores in horrible conditions.

That tub looks so wet they are swimming.  I think your next step is to laugh that one popped out of that, and toss it out.

Was wondering if I could get some info about two newer cube strains called STAKZ and Bluey Vuitton.  I've been able to learn a little bit from other sites but nothing too detailed.  Any info. that could be shared would be greatly appreciated.

They’re cubes so just general cube conditions.

If you’re starting from spores then there’s no way to really predict anything like speed or strength, it’ll just be random each time.

THIS IS MY FIRST TIME WITH MUSHROOMS. I HAVE 9 QUART JARS. 5 MAYBE 6 LOOK LIKE TO ME THAT THEY ARE COMPLETELY COLONIZED AND READY TO GO INTO A BIRTHING CHAMBER. I HAVE ONE READY WITH 2 GALLONS OF PERLITE WASHED AND DRAINED. I WAS GOING TO PUT THEM IN A GREENHOUSE THAT WILL NOT RECEIVE DIRECT SUNLIGHT BUT LOTS OF INDIRECT. THIS IS MY QUESTION SHOULD I CUT THEM IN HALF AND CAN I PUT 2 JARS TOGETHER THAT ARE THE SAME STRAIN. I HAVE 2 JARS THAT HAVE GREEN ON TOP. I'VE RESEARCHED THIS AND SOME SAY IT'S FINE JUST CUT IT OFF OTHERS SAY IT CONTAMINATED WHAT DO YOU THINK.
