You need to wait to grow out a bit before you go to grain.

If you have 4 oz jars, you can just make agar media, pour it into them, and sterilize. Lots of other containers (anything glass or polypropylene) work for makeshift reusable plates you can pour before sterilizing too.

Though if you only have half a cm of growth, it's going to be a long time before you truly need to transfer. Assuming it's clean, it'll be fine for at least a couple of months at room temp. No need to sweat having to wait for supplies.

Wasn't clear from your post whether you are waiting on premade plates or petri dishes to pour into, but I'd point out that if you're not pouring your own plates, you are spending a lot more money than you need to.

Plates I, II, IV and V six days after transfer. III and some of the unnumbered ones had contaminated spots and some had failed to sprout anything at all (I was too dainty with the inoculation loop, I reckon) so I chucked them. Still, these four ought to be enough to get something to transfer into grains.

Hello my beautiful people, so I just wanted to ask what do yall think causes these differences in PE growth on agar and which do you think is the preferred one, for reference, both are using the same high nutrient agar mix (1:1 - LME:AgarAgar).

Yeah, good point, personally it's been pretty hard for me to get a uniform growth of PE on my plates, for example this is a more recent b+ plate, much more uniform growth comparatively. I get that there is no obvious rhizo growth here, but that depends more so on the plate nutrition, no?

I'm running my WWA back to MEA to check for any signs of contamination but I haven't had really any issues, but clearer media is always nice.

Could I please get an experienced eye over these melmac plates? It's from a vendor LC that was riddled with orange bacteria that I've been cleaning up.

I got some premade plates coming. And soon I'm gonna start working with pouring my own agar because yea with how fast I blew thru them 10 first plates I can see where this is going. I can't afford it lol. I dunno if I even got the resources for agae tho unless they come in the mail I live super rural no place here really sells agar shit unless... Bomgaars has it? I just got some more liquid culture in the mail so there's that at least to play with once I'm done with these plates. I'm wondering if the point of failure has something to do with my SAB setup. Its pretty crude. And I'm sterilizing a kitchen knife to do the work. On a sterilized counter surface. I read some stuff recently about keeping the surface you work on covered in like a wet towel and have your dishes set on some grate so that you don't disturb spores and dust on the slick countertop surface and get it moving around in the air your working in. I let the SAB sit 10 minutes after I set up the tools and dishes inside too. But then I think I'm just settling a bunch of trich spores on my counter top just to have them flow into my dishes while I'm workin

The density and patchy growth bother me.  Maybe back the nutes down a tad on your medium to see if it loosens up the culture/coaxes some ropes to get a better isolation of clean growth.  As always, take minimal xfers from the leading edge of growth.
