Hi there, agar peeps! I'm incredibly new to doing Agar work, would love any thoughts on what to look out for on these plates and what to do next (transfer to new plates, set them on fire, etc.,). I know at least one looks contaminated, not sure on the others though. These are all from the same cluster of Z-Strain.

Hey! From my limited knowledge, I can say that:
#4 - Looks like the kind of contam that carries on with the mycelium. Extremely hard to get rid of, I am on my Transfer #7 on my Hawaii MS and it still looks like that.. it is like I can't get rid of it. :sad:

#1 looks similarly contaminated: this kind of contam makes it very hard to see where you could transfer from as it all looks universally crazy.

#2 has a healthy grow at 5'o clock, transfer from there!

#3 looks like the same bizarre contam that is super sticky

#5 looks like it has contam in the middle but healthy grow at the sides, try to transfer from around 5'o clock again!

Good luck! I have loved agar ever since starting in December! :grin:

Disodium Erioglaucine. The dye also contains propylparaben and propylene glycol.

This is 100% circumstantial, so take what I say with a grain of salt, but I've had nothing but problems with Club House brand blue dye. I'll have to check the ingredient list, but it did have PG.

When using it, my cube plates wouldn't grow rhyzo no matter what I did. I tried josex poke, cabin sequestering, and a bunch of transfers. I switched to yellow one day out of boredom and all of a sudden I have rhyzo growth. I now don't use any dye, but it seemed like an interesting coincidence to me.

You can say that again! This is about 36 hours after transfer. You can even see in the original plates where I took the samples from the myc is even starting to grow into the cutout holes! Also the last 4 pics are of some plates I inoculated 12 days ago with spore prints and mss.

And I have to confess, I had to order some premade spawn jars. I am going away for a few days and just don’t have the time to make them this coming weekend. Not happy about it, because I have been having good results making my own jars, but it is what it is.


T1 Transfers at 36 hours and the plates they were taken from…

That little 0.5 cm good enough to go into a 4oz jar of sterile grain? I think I'll wait a bit. I've transferred cultures about 3 times already. I'm out of plates! So I'm like between a rock and a hard place I got no backups till they come in mail so I dunno.. Maybe I need to chance it and transfer it to a grain jar...
