How are these looking now after 13 days? I’m thinking I can start some T1 plates, but where do I look to select what I want to transfer to new plates?

So far I’m happy with what I see, as well as how my first plates that I poured are performing.

Looking good!

Grab the 3 and 9 o clock on the second purple mystic, and the miiddle 6 o clock from the top one.

I prefer the 2nd plates in all of your pictures actually, grab from those. Your first Ochra as well.

you are on the right track, good job!

I tried a thread on charcoal plates, but didn't get any responses on what to transfer, what to send or what to trash. I'll try here.

First the possible send to grains:

Next the possible transfers:

Or are they contaminated and I shouldn't try to save anything?

Not quite sure what the dust is, that is on the lid I hope and not the agar?

Your above plates look good, maybe half of those per jar?

The below plates needs more transfers, but are looking like they have decent growth in multiple places. Those are definitely not for putting to grains, but you could safely take rhizomorphic samples from any of those

Excellent, thanks Huskies. Yeah, was using an insanely bright flashlight and it picks up on all the dust on top of the lid.

I'll give the top ones a try on grain, and transfer the rest.

Thanks! I appreciate it Husk! This is all my first attempt with agar. I had to trash four plates (two pan cyan, two AHB) but these eight I’m pretty excited about. I’ll probably take an 8 or 10mm punch and take a couple samples from each to make T1 plates. Question…What do I do or what is traditionally done with these plates after I take the samples from them?

Hotbox: Cheers, best of luck! Don't forget to take samples from the good plates onto new ones.

Starman: Those might be too big of a sample. Like, it's not an issue, but the larger the sample, the larger the chance of accidentally bringing in contaminants. Some folks here have started taking needle samples.

Sure, it saves you a little bit of time, but since the mycelium growth is exponential, it's better to have a smaller sample.

Usually, extra plates are composted. You can try dehydrating them and make weird dehydrated agar art (if they are not moldy of course).

Don't think of these as waste. Waste is 2kg worth of PCed grains and a brick of coir.

Ahhh ok…good info on all! I bought a punch set that has sizes from 1mm up to 10mm. I can experiment a little I guess.

How many plates do you recommend to transfer to? I am gonna make a shit ton of new plates this weekend.

Compost it is! I’ll put them out to the same place I dump my used up totes. I’m hoping I’ll get an outdoor crop one day there.

I normally do 2 plates, if a plate is very promising and there are multiple strong and distinct areas, you can do more.

Was just working on some transfers (I'm way behind on that shit), and I found some bonus plins!

WGT -from spores collected in my outdoor bed:

PE7 - on a BRF cake that's about ready to get the dunk:

And the bonus PE7 plin(s):

So this plate was kinda shredded up on the surface a bit, then I added 10mL of sterilized H2O, put the lid back on, and shook the ever-living fuck out of it. Sucked up that juice, and promptly spooted it into the quart size BRF cake shown above on March 8th. I put the lid back on the plate so it wouldn't spill any remaining water, then ignored it. Fast forward 3 weeks, and I find it under some other spent plates with 2 plins! The older one looks kinda dry and old. The newer one dropped many spores.

Pretty sure this is painfully obvious, but can someone just confirm this is mold before I toss it all?

Yes it’s a very impressive specimen of mold at that.

Not to be rude, but what about that plate left any doubt in your mind that it was mold? Asking so I can help, not make fun

Totally fair question.

I haven't done this type of Pan before so I was holding out a tiny hope that it was some kind of mycelium variation I was ignorant of. 😔 Also I know it wasn't my sterile technique because all my other plates and LC from that day (including agar transfer, a clone, and another LC syringe from the same trusted vendor) are all fine. And I was thinking, well if it's contam it's seriously contaminated. How likely is that?

And I have some from this syringe in a jar of LC and the mycelium is growing fantastic. So I really didn't want to toss everything and learn later that it was fine. TBH I guess in my heart I knew it was toast I just didn't want to admit it or belive my own eyes. 🙃  lesson learned: be less attached to projects that have obviously failed.

Thanks to both of you for the confirmation.

ETA: Oh and it's 3 different plates :facepalm3:

In better and less embarrassing news, the first clone I put on agar is doing nicely:

The clone is from 3/21. The mold is from Sunday.

Was that a LC syringe from a trusted vendor? PanCyan Peace River by any chance?

The mold one was exactly that, yes.

Is the yellow pigment in the agar intentional or did the mold cause that to happen?
