Those ^^^ secondary growth spots were me moving the plug around to position it in the middle and face down… I’ve since decided I’m not going to worry about going face down and just drop the plug from the punch where I want it.  It doesn’t seem to take the myc long to grow down to the plate anyway and it’s not worth the hassle and possibly flinging the plug off the plate which has almost happened a couple times lol



Swab Streaked MiB.  I had a print (thanks again Mojave!—that shit was clean, Son.. at least to my untrained eye), used a set of swabs.. put a couple drops of sterile water on one swab and streaked the print once.  Then added another drop and used the second swab to rub the spore swab to get the spores all nice and loose and in the water on the tip, then streaked the plate





This one I used a cell spreader on.. obv used a little too much water as it ended up pooling on the edge of the plate.. love and learn



Just kinda fackin around testing the limits of this media.. I noticed in one of my plates I transfered away from at the leading edge that I cut a little long on the edge side of the triangle and where my cut ended, it had myc growing there.  Like I dragged myc away from the edge out into the open agar.  Thinking I could maybe use this in some instances to just pull myc away if I’m trying to clean up bacterial myc without having to use another plate and do it really quickly. 

Trying to replicate it on one of these clean plates.. at first I thought that using the blunt side of the scalpel blade would work better for dragging.. as you can see, it wasn’t enjoyable or easy :lol:  so tried it with the sharp side on the left.  Way easier and more accurate to get juuust on the edge.  We’ll see what happens.  Again, just playin around.

I have tried blagar twice now and have gotten the stuff you see in about half of the plates I pour of it. I don't have this issue with not using activated charcoal. I don't know why it keeps happening. If I put a stir bar in it and maybe stirred while it was in sab for little while on magnetic stirrer Will that maybe help get them out?

even with tissue cloning,  I employ the "drag" method.

It amazing how just a little touch to the media can be successful & clean.  Even when the sample it's self doesn't turn out as clean.

I feel like we're about to get some major break throughs, here soon, in the agar world. Now that faht has caught the bug.

What brand activated charcoal do you use? I use Anthony's Activated Charcoal and stir with a stick blender. I heat it on the stove on medium low heat and mix while heating.

Then I pour it into pint jars to PC, and stir the pot every jar or two of pouring.

That doesn't look like contam, it looks like harmless air bubbles. I would think your issue is more likely not stirring enough more than the brand of charcoal. (or maybe stirring TOO much, if stirring with magnetic stirrer on highest setting? ...except that you don't have the issue without the charcoal.

Whatever the issue is it looks like your agar gelled while still having bubbles on top. I have never seen bubbles on mine.

Took some early transfers from this but after growing out this plate looks the best and appears clean. Is it safe to put to grain?

God awful pics, but it was best I could do with HGs and condensation. Myc is real thick and raised up...I'm not seeing anything "typical" that I could grab from. I have other plates of it growing, but I was curious to know if this was indicative of anything in particular. It's x7x+, if it matters.

I have been pouring them thick lately - the depth of the HG plates make it a bit of a PITA sometimes, so I poured the last batch thicker.

I believe the nutes were still 2%. I may have reduced the agar from 2% to 1.7% though. I did with my last round of plates but I can't remember if I did with the batch that this plate came from.

There's also a chance that I smothered this one with having the lid on tight. Idk if that would cause this appearance though.

I was only asking because I’ve had experiences with cultures going super tomentose with thick plates or high nutes. I try to keep my plates relatively thin, but not super thin. Like maybe an eighth inch, or just a cunt hair thinner. Just to kind of keep it standard, which makes odd growth much more apparent

Puffy and raised up can mean bacterial, but occasionally not.

Do the rest of them look better? If not I'd grab from the very edge of this at 4 o'clock. Looks less puffy there and maybe hinting at rhizo. 

Plates can be poured much thinner than that in the future.

I know x7x+ is usually rhizo at all phases but the one exception in my experience is on germination plates. Here's a typical clean x7x+ germination plate of mine.

Tbh, I'm straight up dumb as shit with a camera all around, and I mostly wanted to capture the raised behavior, which felt good enough to get the point. Lmao I poured thick to reduce the angle I'd work at bc the HGs are kinda deep. Maybe that was a bad idea I didnt anticipate. I was also shooting for plate longevity while I wait for an opportunity to buy cryovials. Either way, message received - thank you.

I’ve just recently started working with agar.

I poured some plates and made transfers and I now have quite a few plates that have grown to the edge. Here is an example of what I mean…






I’m not ready to use these plates quite yet. What do I do with them?  Can I put them in the refrigerator?  About how long do I have to use them before they go bad?  I’m new at this. I’m afraid I may have made too many plates not knowing any better. I didn’t put much thought into how long a colonized agar plate is viable.

Any advise is appreciated.
I’ve got a lot more than just those pictures. Some have started climbing the sides and I’m afraid if I can’t slow it down it’ll find a way to contaminate itself or die or I don’t know. Something bad rust wastes my time and effort I’m sure. Ha.


Is there a way to long term store these or in the future only make one at a time?

Wrap, bag and refridgerator. I have yet to test the limits, but 6 months with cubes ok by doing it like that.
Stasis in sterile water or slants will work well.

Bag it for sure... fridge if you can guarantee it won't freeze AND need to slow it down.

I've got plenty from a good 9-12+months plates that just got a hot pour "feeding". And they be looking 👌. Also remember a little more nutes or additional (nutritional yeast is a fav) will keep the myc happy longer. But won't (necessarily) be all pretty rhizzy like that, actually though they could . Just won't be running out the door so quickly.
